Abstract The precise mode of action ganaplacide (KAF156), a phase III antimalarial candidate, remains elusive. Here we employ omics-based methods with the closely related chemical analog, GNF179, to search for potential Plasmodium targets. Ranking targets derived from genetics and proteomic affinity chromatography methodologies identifies SEY1, or Synthetic Enhancement YOP1, which is predicted encode an essential dynamin-like GTPase implicated in homotypic fusion endoplasmic reticulum (ER) membranes. We demonstrate that GNF179 decreases SEY1 melting temperature. further show binds recombinant subsequently inhibits its activity, required maintaining ER architecture. Using ultrastructure expansion microscopy, find treatment changes parasite Golgi morphology. also confirm SEY1 gene P. falciparum. Zika and malaria are important vector-borne febrile illnesses in humans. In this study, we determined the circulation of virus infections, their hotspots, predominant clinical features. A cross-sectional study was carried out six Local Government Areas (LGAs) Kaduna State, Nigeria, from September 2018 to May 2019. Four hundred twenty sera were screened for (ZV) IgM IgG, Plasmodium falciparum antigen using ELISA immunochromatographic test, respectively. Overall, a seroprevalence 14.5% found Zika, 9.3% malaria. Nineteen (4.5%) thirty-five (8.3%) patients seropositive ZV Co-infection rates (ZV IgM) (0.5%: 2/420), IgG (0.7%: 3/420) observed. Lere (10%: 7/70 IgM), Kachia (14.3%: 10/70 IgG) Zaria (18.6%: 13/70 malaria) LGAs identified as hotspots Age significantly associated with (p = 0.008) 0.004). Patients aged 1–10 years had highest (18.4%), while those 21–30 prevalence (6.1%: 7/114). Ring hemorrhages are characteristic ring-shaped lesions in the parenchyma of central nervous system (CNS) surrounding small blood vessels and typically reported to occur white matter brain. In humans, they seen various diseases, including pernicious anemia, cerebral fat embolism, malaria. also sporadically CNS animals have been previously 4 nonhuman primate (NHP) species with forms anemia. Here we present results preclinical toxicity studies testing drug candidates different modalities, targets, mechanisms action, indications NHPs (cynomolgus monkeys). Within each study, ring were observed associated severe anemia affected line those reported. They occurred gray occasionally brain spinal cord; brain, thalamus, basal ganglia, cerebellum particularly affected. Lesions comprised a eosinophilic core, sometimes or slightly basophilic amorphous material center, surrounded by red cells and/or microglial microglia aggregates. Fibrin staining confirmed presence fibrin core. No commonality type/cause was noted; one considered spontaneous (non-treatment-related) finding.
