Polymorphonuclear lymphocytes (PMNs) are terminally differentiated phagocytes with pivotal roles in infection, inflammation, tissue injury, and resolution. PMNs can display a breadth of responses to diverse endogenous exogenous stimuli, making understanding these innate immune responders vital yet challenging achieve. Here, we report 22-color spectral flow cytometry panel profile primary human on population single cell levels for surface marker expression activation, degranulation, phagocytosis, migration, chemotaxis, interaction fluorescently labeled cargo. We demonstrate the protein response phorbol ester stimulation compared untreated controls an adherent PMN model additional analysis intra- inter-subject variability. challenged Gram-negative bacterial pathogen

Abstract Liquid biopsies, such as a blood sample, are non-invasive and simple ways to monitor biomarkers in clinical settings. For cancer immunotherapy settings major goal is identify host immunological that can be used progression of disease, treatment efficacy, or even adverse effects. Here, the profiling validation circulating immune cells may allow continuously survey on how system alters during disease progress treatment. However, practice will require proper sampling development robust high-dimensional phenotypic analysis. we designed 37-marker spectral flow cytometry assay with purpose conducting immunophenotyping monitoring status patients. The was developed profile all cell compartments, their differentiation well activation markers, including numerous checkpoint markers. Importantly, validated this context liquid biopsies should processed ensure minimal effect cryopreservation compared freshly acquired sample. Our work highlights important considerations for which alternations introduced different methods. Essentially, identified fast processing pipeline where showed changes profile. This approach certainly implemented help diagnostic sites strong tool applicable individual patients’ status.
