Due
to
the
high
reproduction
rate
of
COVID-19,
it
is
important
identify
and
isolate
infected
patients
at
early
stages
infection.
The
limitations
current
diagnostic
methods
are
speed,
cost,
accuracy.
Furthermore,
new
viral
variants
have
emerged
with
higher
rates
infectivity
mortality,
many
mutations
various
primer
binding
sites,
which
may
evade
detection
via
conventional
PCR
kits.
Therefore,
a
rapid
method
that
sensitive,
specific,
cost-effective
needed
for
point-of-care
molecular
test.
Accordingly,
we
developed
SARS-CoV-2
kit
specificity
sensitivity,
RT-PCR,
taking
advantage
loop-mediated
isothermal
amplification
(LAMP)
technique.
Four
sets
six
primers
were
designed
based
on
conserved
regions
genome:
two
outer,
inner
loop
primers.
Using
optimized
protocol,
genes
detected
as
quickly
10
min
but
most
sensitive
30
min,
detecting
little
100
copies
template
DNA.
We
then
coupled
RT-LAMP
lateral
flow
dipstick
(LFD)
multiplex
detection.
LFD
could
detect
genic
amplifications
single
strip,
making
suitable
multiplexed
development
RT-LAMP-LFD
reaction
crude
VTM
samples
would
be
diagnosis
COVID-19
in
laboratories
well
private
homes.

designed
targeting
the
mitochondrial
cytochrome
C
oxidase
I
(COI)
gene.
assay
was
able
detect
low
820
fg/L
of
DNA
in
60
min
at
65
°C,
hundredfold
higher
than
detection
limit
classical
polymerase
chain
did
not
show
cross-reactivity.
conclusion,
LAMP
demonstrated
excellent
sensitivity
byproducts,
sampling-to-result
