Giardiasis is a parasitic disease caused by Giardia lamblia (G. lamblia) that affects people worldwide. Still, few studies report on the immunoregulatory effects of biomolecules colostrum during interactions with G. lamblia. This study aimed to assess concentrations melatonin and cortisol hormones, percentage Treg cells, levels cytokines IL-10 TGF-β in from mothers who tested positive for parasite. cross-sectional analyzed samples 25 puerperal. The were using an ELISA determine if they seropositive type antibody present (IgM IgG). Based results, divided into three groups: control group (N = 10) no reaction either IgM or IgG, IgG (IgG+/IgM−; N 8), (IgM+/IgG−; 7). measured method. Additionally, immunophenotyping flow cytometry. In anti-G. lamblia, concentration was lower. However, level this hormone had increased. similar between groups, regardless seropositivity. There higher cells also increased seronegative there correlation cells. These data suggest increase regulatory reduction recent giardia infection may contribute evolution manifestation disease.

The pharmacological treatment of Giardia lamblia infection involves the use chemical agents, such as metronidazole (MNZ). However, these medications are associated with a range adverse effects, and their effectiveness is not definitively established. In light previously discussed information recognized antimicrobial properties Ferula macrecolea, this study aims to investigate both in vitro vivo anti-giardial effects F. macrecolea essential oil (FME) on G. infection. Gas chromatography-mass spectrometry (GC-MS) was utilized analyze composition prepared FME. MTT colorimetric assay employed assess FME's cytotoxic activities. were evaluated compared MNZ mice infected lamblia. Additionally, FME therapy serum electrolyte levels expression inflammatory cytokines assessed. primary components identified terpinolene (78.72%), n-nonanal (4.47%), linalool (4.35%). significantly reduced viability growth rate trophozoites (IC50 = 21.6 μg/mL) cysts 27.6 dose-dependent manner control group (p < 0.001). CC50 value for against normal intestinal cells determined be 207.4 μg/mL. vivo, assays demonstrated that administration various doses FME, particularly combination over 7 days, resulted statistically significant reduction mean number cysts, level electrolytes (sodium potassium), interleukin-1 (IL-1), tumor necrosis factor-alpha (TNF-α), nuclear factor κB p65 (NF-κB p65), Toll-like receptor 4 (TLR-4) giardiasis This study's results demonstrate extract's efficacy lamblia, exhibiting minimal cytotoxicity towards cells. Furthermore, extract shown manage murine models by modulating responses via suppressing NF-κB p65/TLR pathways. further research necessary clarify specific mechanisms action combating
