Click chemistry has become one of the most powerful construction tools in field organic chemistry, materials science, and polymer as it offers hassle-free platforms for high-yielding synthesis novel easy functionalization strategies. The absence harsh reaction conditions or complicated workup procedures allowed rapid development biofunctional polymeric materials, such biopolymers, tailor-made surfaces, stimulus-responsive polymers, etc. In this review, we discuss various types click reactions─including azide-alkyne cycloadditions, nucleophilic radical thiol reactions, a range cycloadditions (Diels-Alder, tetrazole, nitrile oxide, etc.), sulfur fluoride exchange (SuFEx) reaction, oxime-hydrazone reactions─and their use formation study polymers. Following that, state-of-the-art biological applications "click"-biofunctionalized including both passive (e.g., biosensing bioimaging) "active" ones that aim to direct changes biosystems, e.g., drug delivery, antiviral action, tissue engineering. conclusion, have outlined future directions existing challenges click-based polymers medicinal clinical applications.

Injuries to the central nervous system (CNS) often lead persistent inflammation and limited regeneration. This study developed a clinically relevant injectable hydrogel derived from decellularized human peripheral nerves, with mechanical properties biomimicking native CNS tissue. Using modified Hudson method, sciatic nerves were decellularized, effectively removing immunogenic cellular debris while retaining extracellular matrix. Two delipidation solvents, dichloromethane: ethanol (2:1 v/v) n-hexane: isopropanol (3:1 v/v), evaluated, former achieving optimal lipid removal better digestion. The resulting solution was crosslinked genipin, forming an (iHPN) that gelled within 12 min at 37 °C exhibited stiffness of approximately 400 Pa. Human astrocytes, microglial cell clone 3 (HMC3), mouse RAW 264.7 macrophages cultured individually iHPN, lipopolysaccharide (LPS) added mimic following injury. Compared LPS-activated cells on tissue culture plates (TCP), astrocytes iHPN maintained quiescent state, as evidenced by reduced GFAP expression IL-1β secretion. HMC3 in displayed anti-inflammatory phenotype, shown increased CD206 decreased CD86/CD68 expression, along higher IL-4 lower TNF-α/IL-1β SH-SY5Y neuroblastoma viability improved neuronal differentiation compared TCP. brain neurons had TCP or collagen hydrogels. Overall, is novel has potential for minimally invasive applications, such carrier drug delivery and/or biomaterial support axonal growth.

Regeneration of the injured endometrium, particularly functional layer, is crucial for prevention uterine infertility. At present, clinical treatment using sodium hyaluronate hydrogel injection limited by its relatively low fluidity, short-term retention, and insufficient bioactive ingredients, so it necessary to develop an advanced healing-promoting hydrogel. The modulation microenvironment Lactobacillus presents a component that can facilitate regeneration layer. Our study introduces multifunctional Lactobacillus-loaded poly(N-isopropylacrylamide)-grafted bacterial cellulose (BC-g-PN@L) designed with superior injectability in situ stability. 25 °C (room temperature), uniform distribution achieved pressure only 7.90 kPa. 37 (body BC-g-PN@L forms robust three-dimensional nanonetwork, providing space substance exchange channels maintain viability bioactivity. Enhanced hydrophobic isopropyl groups poly(N-isopropylacrylamide) side chains rigid substrates, exhibits prolonged retention properties cavity, persisting over 21 days. These attributes endow versatile pro-healing capacity rat model endometrial injury. promotes development injectable hydrogels both histological repair endometrium.

Severe endometrial injuries may cause thin endometrium and intrauterine adhesion in women which can result uterine factor infertility. Current treatments, including surgical separation of adhesions hormonal regeneration the endometrium, often fail to prevent re-adhesion achieve satisfactory reproductive results. Recently, mesenchymal stem cells (MSCs) have become a promising new treatment for IUA. However, challenges such as cell survival transplantation limit effectiveness MSC therapy. Researchers explored various approaches enhance therapeutic efficiency MSCs. Among these, biomaterials been frequently employed due their biocompatibility, degradability, ability provide conducive environment growth. This review discusses use MSC-based therapies reconstruction summarizes evidence from preclinical clinical studies, highlighting efficacy safety these biomaterials. The also addresses future directions this field, advances biomaterial engineering, currently under investigation, personalized medicine approaches. emphasizes significance therapy provides practical guidance developing materials protocols applications.

Mesenchymal stem cells (MSCs) are a source of wide range soluble factors, including different proteins, growth cytokines, chemokines, and DNA RNA molecules, in addition to numerous secondary metabolites byproducts their metabolism. MSC secretome can be formally divided into secretory vesicular parts, both which very important for intercellular communication involved processes such as angiogenesis, proliferation, immunomodulation. Exosomes thought have the same content function MSCs from they derived, but also number advantages over cells, low immunogenicity, unaltered functional activity during freezing thawing, lack tumor formation. In addition, pre-treatment with various inflammatory factors or hypoxia alter secretomes so that it modified more effective treatment. Paracrine secreted by improve survival other cell populations several mechanisms, immunomodulatory (mostly anti-inflammatory) anti-apoptotic partly based on Hsp27 upregulation.
