The clinical application of human umbilical cord mesenchymal stem cells (hUCMSCs) in the treatment liver failure faces challenges due to cell quality, short engraftment time, and limited efficacy. Here, gelatin microcryogel (GM) microcarriers with pore sizes ranging from 15 36 μm were tuned mixed glutaraldehyde develop a 3D culture system hUCMSCs improved therapeutic effects. Bulk RNA sequencing vitro assays showed that 3D-hUCMSCs exhibited significant improvement signaling pathways related paracrine secretion anti-inflammation. These superior compared 2D-hUCMSCs not only terms secretion, protection oxidation, resistance mechanical force damage, but also had better function effect than when they transplanted as single into injury mice. Furthermore, sponge patch grafting (GSPG) strategy was developed enable direct within GM microcarriers. results overall host significantly improved, life span hosts extended. Our study provided practical achieve high long retraining time rescuing acute matrixes.
