Neuropeptide Y (NPY) is a 36-amino acid neuropeptide that widely expressed in the central nervous system, including nucleus accumbens (NAc), hypothalamus, and amygdala. The NAc involved several behaviors, reward, motivation processes, feeding behavior. Here, we demonstrate male mice NPY input from basolateral amygdala (BLA) to preferential consumption of high-fat diet (HFD). First, demonstrated BLA by injecting adeno-associated virus (AAV)(retro)-FLEX-mCherry into NPY-Cre mice. We also confirmed neurons project exclusively AAV(dj)-hSyn-FLEx -mGFP-2A-Synaptophysin-mRuby BLA. Usually, HFD drives enhanced food intake than standard chow after repetitive exposure. optogenetic inactivation projecting caused significant decrease for 1-h period, while activation these induced opposite effect. Furthermore, bilateral injection an receptor type 1 (Y1R) antagonist significantly decreased period compared with vehicle injection, while, conversely, Y1R agonist intake. These results suggest mediate via NAc-localized Y1R.
