Polyphenols constitute an important group of natural products that are traditionally associated with a wide range bioactivities. These usually found in low concentrations and now available nutraceuticals or dietary supplements. A polyphenols include apigenin, quercetin, curcumin, resveratrol, EGCG, kaempferol have been shown to regulate signaling pathways central for cancer development, progression, metastasis. Here, we describe novel mechanistic insights on the effect this key elements impacting cancer. We protein modifications induced by these their several including PI3K, Akt, mTOR, RAS, MAPK particularly those affecting tumor suppressor p53 protein. Modifications gene expression levels posttranslational such as phosphorylation, acetylation, ubiquitination influence stability, subcellular location, activation new transcriptional targets, role response DNA damage, apoptosis control, cell- cycle regulation, senescence, cell fate. Thus, deep understanding effects players cancer-driving will certainly lead better designed targeted therapies, less toxicity treatment. The scope review centers regulation most studied highlights importance profound regulations order improve treatment control products.

Understanding the mechanisms of food digestion is paramount importance to determine effect foods have on human health. Significant knowledge fate during has been generated in healthy adults due development physiologically-relevant vitro models. However, it appears that performance oro-gastrointestinal tract affected by ageing and a model simulating digestive conditions found younger adult (<65 years) not relevant for an older (>65 years). The objectives present paper were: (1) conduct exhaustive literature search find data physiological parameters tract, (2) define adapted adult. International experts discussed all dedicated workshop organized within INFOGEST network. Data bolus properties collected were gathered, including particle size boluses. In stomach small intestine, suggest significant changes are observed between adults. latter, rate gastric emptying slowed down, pH content higher, amount secretions thus hydrolytic activities intestinal enzymes reduced concentration bile salts lower. consensus proposed here will allow progress be made understanding this specific population, facilitating their nutritional needs. Nevertheless, better foundational when available further refinement needed implement future.

Natural products have been extensively applied in clinical practice, characterized by multi-component and multi-target, many pharmacodynamic substances, complex action mechanisms, various physiological activities. For the oral administration of natural products, gut microbiota efficacy are closely related, but this relationship remains unclear. Gut microbes play an important role transformation utilization caused diversity enzyme systems. Effective components such as flavonoids, alkaloids, lignans, phenols cannot be metabolized directly through human digestive enzymes can transformed produced microorganisms then utilized. Therefore, focus is paid to metabolism microbiota. In present study, we systematically reviewed studies about their effect on biotransformation highlighted involved common bacteria, reaction types, pharmacological actions, research methods. This study aims provide theoretical support for application prevention treatment diseases new ideas studying based biotransformation.

As a subclass of the biopharmaceutical classification system (BCS) class II, basic drugs (BCS IIB) exhibit pH-dependent solubility and tend to generate supersaturation in gastrointestinal tract, leading less qualified vitro-in vivo correlation (IVIVC). This study aims develop physiologically based multi-cup dissolution approach improve evaluation for higher quality IVIVC preliminarily explores molecular mechanism precipitation ketoconazole affected by Polyvinylpyrrolidone-vinyl acetate copolymer (PVPVA) hydroxypropyl methyl-cellulose (HPMC). The concentration each cup dynamic model (DGIM) was measured using fiber optical probes. Molecular interactions between PVPVA or HPMC were simulated Materials Studio. results demonstrated that improved maintained ketoconazole. exhibited superior inhibitory effect on molecule aggregation due slightly stronger van der Waals forces as well unique electrostatic forces, thereby further enhancing vitro drug absorption, which correlated with absorption. Compared conventional apparatus paddle method, DGIM mean prediction error through from 19.30% 9.96%, reaching qualification criteria. In conclusion, enables transportation BCS IIB ketoconazole, enabling screening screen inhibitors achieving formulation studies.
