The free expansion of a confined chain in theta solvents following sudden removal the confining constraint is investigated using Langevin dynamics simulations both two- and three-dimensional spaces. average evolution size exhibits sigmoidal transition between states on logarithmic timescale, indicating two-stage expansion, each characterized by its own timescale. A kinetic theory developed applying Onsager's variational principle, which balances change energy with dissipation. Through scaling analysis, characteristic time τ1 for first stage shown to scale as cube initial size, while increases according power law an exponent α1=1/3 , independent spatial dimension. In second stage, timescale τ2 found be proportional square length, follows exponential recovery function powered α2=1/4 . These results are further validated direct analysis equations via simulations. Moreover, general forms two stages established through integration equations. Finally, physical interpretations proposed, employing radial model diffusive mechanism explain observed behaviors. This work explores system under specific solvent condition, providing foundational enhancing our understanding expansion-upon-release phenomenon.

mTORopathies represent a group of neurodevelopmental disorders linked to dysregulated mTOR signaling, resulting in conditions such as tuberous sclerosis complex, focal cortical dysplasia, hemimegalencephaly, and Smith–Kingsmore Syndrome. These often manifest with epilepsy, cognitive impairments, and, some cases, structural brain anomalies. The pathway, central regulator cell growth metabolism, plays crucial role development, where its hyperactivation leads abnormal neuroplasticity, tumor formation, heightened neuronal excitability. Current treatments primarily rely on inhibitors, rapamycin, which reduce seizure frequency size but fail address underlying genetic causes. Advances gene editing, particularly via CRISPR/Cas9, offer promising avenues for precision therapies targeting the mutations driving mTORopathies. New delivery systems, including viral non-viral vectors, aim enhance specificity efficacy these therapies, potentially transforming management disorders.
