Chemotherapy plays a crucial role in triple-negative breast cancer (TNBC) treatment as it not only directly kills cells but also induces immunogenic cell death. However, the chemotherapeutic efficacy was strongly restricted by acidic and hypoxic tumor environment. Herein, we have successfully formulated PLGA-based nanoparticles concurrently loaded with doxorubicin (DOX), hemoglobin (Hb) CaCO3 CaCO3-assisted emulsion method, aiming at effective of TNBC. We found that obtained nanomedicine (DHCaNPs) exhibited drug encapsulation pH-responsive release behavior. Moreover, DHCaNPs demonstrated robust capabilities neutralizing protons oxygen transport. Consequently, could serve nanoshuttles to attenuate hypoxia neutralize microenvironment (TME) depleting lactic acid, thereby effectively overcoming resistance chemotherapy. Furthermore, notable ability enhance antitumor immune responses increasing frequency tumor-infiltrating effector lymphocytes reducing various immune-suppressive cells, therefore exhibiting superior suppressing growth metastasis when combined anti-PD-L1 (αPD-L1) immunotherapy. In summary, this study highlights TME, offering promising strategy figure out an enhanced chemo-immunotherapy benefit TNBC patients.

Chemoimmunotherapy has the potential to enhance chemotherapy and modulate immunosuppressive tumor microenvironment by activating immunogenic cell death (ICD), making it a promising strategy for clinical application. Alantolactone (A) was found augment anticancer efficacy of paclitaxel (P) at molar ratio 1:0.5 (P:A) through induction more potent ICD via modulation STAT3 signaling pathways. Nano drug delivery systems can synergistically combine natural drugs with conventional chemotherapeutic agents, thereby enhancing multi-drug chemoimmunotherapy. To improve targeting ability bioavailability hydrophobic drugs, an amphiphilic prodrug conjugate (HA-PTX) chemically modified (PTX) hyaluronic acid (HA) as backbone.
