In the present study, three experiments were designed to identify most appropriate technique for freezing rabbit semen. Experiment 1 aimed determine optimal levels of dimethyl sulfoxide (DMSO) contents in medium and their effects on individual bucks. Semen ejaculates each buck (n=15 bucks) mixed split into portions extension with a containing varying concentrations DMSO (0.75, 1.0, 1.4 M). Diluted semen samples packaged 0.25 mL straws suspended above liquid nitrogen (LN) 10 min, then dipped LN. Reproductive technologies, including sperm cryopreservation, offer conservationists enhanced capacity to genetically manage populations and improve the outcomes of conservation breeding programs (CBPs). Despite this potential, post-thaw quality amphibian is highly variable following research focused on protocol refinement needed. The aim study was twofold: (1) investigate effect addition bovine serum albumin (BSA) cryopreservation medium (pre-freeze), (2) caffeine activation (post-thaw), characteristics in critically endangered Booroolong frog (Litoria booroolongensis). Given the extensive use of plastic materials in modern society, there is an escalating concern about potential risks associated with exposure to products. This study investigated impact leachates from boiled-water-treated cups, including polypropylene (PP), high-density polyethylene (HDPE), low-density (LDPE), polystyrene (PS), and terephthalate (PET), on male reproductive health. Experimental mice were administered daily doses for 180 consecutive days. Histological analysis testes epididymis was conducted, revealing vacuolization absence sperms seminiferous tubules treated PP, HDPE, LDPE, PS, while PET exhibited lower toxicity. In this study, it was hypothesized that the addition of an appropriate concentration Y-27632 (a ROCK inhibitor) to freezing extender prevents cryopreservation-induced apoptosis and improves embryonic development after in vitro fertilization (IVF). Semen samples were collected from five fertile Simmental bulls using artificial vagina twice a week for 4 weeks. Selected pooled diluted with Tris-egg-yolk-glycerol (TEYG) containing different concentrations (0, 10, 20, 30, 40 μM) then frozen liquid nitrogen. After thawing, computer-assisted semen analysis (CASA), plasma membrane integrity, acrosome intactness evaluated terms morphological abnormalities, intracellular generation reactive oxygen species (ROS), DNA fragmentation, phosphatidylserine (PS) externalization, apoptotic-related gene expression.
