Abstract Semen cooling and cryopreservation have revolutionized the field of reproductive biotechnologies. However, challenges persist in maintaining sperm quality viability during these processes. The unsatisfactory results main problems are associated with low quality, viability, morphological, structural, DNA integrity, changes plasma membrane, ability to interact female tract decrease fertilization potential. objective this study was determine effect seminal proteins different molecular weights on kinetic parameters dog spermatozoa cold storage at 4 °C. were isolated from dogs’ ranging between 6-200 kDa, separated four fractions. ejaculates (n=15) collected 6 heathy dogs which pooled. Spermatozoa extended Tris medium alone (control) or addition one protein fractions, then incubated °C for 2 hours. Sperm 10-15 kDa had significantly higher total motility (37.01±2.98%), progressiveness (15.97±1.91%), curvilinear line velocity (37.46±3.75 μm/s), linearity (26.18±1.00%), straightness (45.94±2.03%) compared other groups weight control group. findings indicated that use certain can be beneficial reducing detrimental preserving spermatozoa. It seemed presence canine rendered less amenable negative influence cooling.

Despite decades of research and handling semen for use in artificial insemination (AI) other assisted reproductive technologies, 5-10% selected boar sires are still considered sub-fertile, escaping current assessment methods sperm quality resilience to preservation. As end-product, the ejaculate (emitted spermatozoa sequentially exposed composite seminal plasma, SP) ought define homeostasis testes, epididymis, accessory sexual glands. Yet, linking findings production biology fertility is suboptimal. The present essay critically reviews how a fertile can help us diagnose both health handling, focusing on -available under development- identify suitable biomarkers cryotolerance fertility. Bulk SP, proteins microRNAs (miRNAs) have, albeit linked function after AI, failed enhance outcomes at commercial level, perhaps just being components complex functional pathway. Hence, focus now interaction sperm-SP, comparing vivo with ex vivo, regarding nano-sized lipid bilayer extracellular vesicles (sEVs) as priority. sEVs transport fragile molecules (lipids, proteins, nucleic acids) which, shielded from degradation, mediate cell-to-cell communication female internal genital tract. Such modulates essential processes, immunological tolerance. be harvested, characterized, stored, manipulated, e.g. used andrological diagnosis, selection breeders, alternatively additives improve cryosurvival

Seminal plasma (SP) is not a pre-requisite for pregnancy. Yet, this heterogeneous, composite SP has proven relevant fertility, as mediator cell-to-cell communication between producing cells, spermatozoa and the female internal genital tract, regulating complex reproductive processes. Bearing hormones, proteins, cytokines well nuclei acids in nano-sized lipid bilayer seminal extracellular vesicles (sEVs), concerts signaling to female. Signals influence timing of ovulation, sperm transport and, particularly, enable immune system balance her cryptic choice engage into pregnancy or reject an eventual fertilization. This essay, focusing on livestock general pigs particular, discusses intrinsic roles sEVs with regards homeostasis, while binding internalizing their cargo tract epithelia regulate functional activity. Since prior studies had inconclusive results using bulk single SP-contained free molecules, argumentation hereby provided increase current incipient research sEVs, where fragile molecules fertility are shielded from degradation during handling. EVs isolated can be used andrological diagnosis perhaps select breeders optimal fertility. Moreover, laboratory-uploaded specific even engineered nanodroplets additives extenders improve after artificial insemination (AI) biotechnologies.

Context Collecting, cooling, and cryopreserving semen is essential for accessing genetically superior stallions. However, preserving stallion sperm presents unique challenges compared with other species. Aims This study aimed to investigate the effect of zinc (Zn) supplementation on quality equine during cold storage. Methods Various factors contributing were assessed at 24 48 h after subjecting samples a heat-resistance test. In Experiment 1, four experimental groups examined, each different concentrations Zn sulfate, as follows: 0, 2, 3 mM. Subsequently, 2 involved testing wider range (0, 0.1, 0.2, 0.4, 0.8, 1.6, 3.2 mM), including evaluation incubation 240 min 37°C (heat-resistance test). Key results The addition extender INRA96 did not yield substantial improvements in sperm-quality parameters cooling or h. Moreover, no protective benefits observed when underwent Concentrations surpassing mM had an adverse parameters. Conclusions implications These findings have contributed understanding strategy improving preservation

The present study was conducted to observe the effects of removal seminal plasma Pantja buck semen and supplementation bovine (BSP) in extender before cryopreservation. In a preliminary experiment, different levels BSP were supplemented (1, 3, 5, 7, 9% v/v) egg yolk (7.5% yolk)-tris (EYT) used for cryopreservation semen. Results terms motility, viability, membrane integrity, acrosome lipid peroxidation showed that 5% suitable maintaining quality during final pooled from four bucks split into three aliquots (I, II, III). Aliquot I directly diluted EYT grouped as control (C); aliquot II III washed separately with TALP solution D1 (Washed extender) D2 containing BSP), respectively. Seminal attributes (sperm individual total morphological abnormalities) assessed at postdilution, postequilibration, post-thawing stages. Alanine aminotransferase (ALT), aspartate (AST), malondialdehyde (MDA) concentration, glutathione peroxidase (GSH-Px) activity measured post-thaw. Washed significantly improved (p < 0.05) parameters post-thaw compared unwashed (control). A significant difference observed between freezing stages dilution groups. Significantly higher sperm GSH-Px activity, lower MDA concentration extracellular release enzymes (ALT, AST) group D1. results demonstrated BSP-supplemented can improve quality.
