Background: Neuropathic pain can be triggered by chemotherapy drugs such as paclitaxel (PTX). Management of is limited drugs' ineffectiveness and adverse effects. Isopulegol (ISO) a monoterpene present in the essential oils several aromatic plants has promising pharmacological activities. Objectives: to evaluate antinociceptive activity ISO PTX-induced neuropathic model. Methods: toxicity was evaluated healthy cancerous cells. Behavioral assessments were performed using von Frey acetone tests. We investigated involvement GABAergic pathway, NMDA, TNF-α, release GABA glutamate presence ISO. Results: showed little or no cytotoxicity U87 MDA-MB-231 In both acute subacute treatment, at doses 25, 50, 100 mg/kg (* p < 0.05) increased mechanical nociceptive threshold animals compared control group reduced thermal sensitivity. Its action reversed pre-treatment with flumazenil potentiated NMDA antagonist, MK-801. TNF-α levels tests carried out. Conclusions: shows low neuronal cells its association PTX generated synergism cytotoxic action. The effect due activation antagonism receptors involves stabilization plasma membranes leading an imbalance neurotransmitters, favoring GABA-mediated inhibition over glutamatergic excitation.

Peripherally Inserted Central Catheters (PICCs) and midline catheters are crucial for chemotherapy supportive care in cancer patients. Their use requires ongoing monitoring to prevent late complications such as infections, dislodgements, replacements. The COVID-19 pandemic challenged healthcare systems, potentially increasing these due reduced outpatient services limited specialized personnel. This study compared the incidence of associated with PICCs patients during after pandemic. A retrospective observational was conducted at a Cancer Center Italy from March 2020 April 2024. Catheter-related were divided into two cohorts: (March 2020-March 2022) post-pandemic (April 2022-April 2024). primary outcome requiring device removal, categorized Statistical analyses included Chi-squared test categorical variables Kruskal-Wallis continuous variables. Of 4104 PICC catheter placements, 2291 removals recorded, 550 (24%) complications-404 146 (p < 0.001). Suspected infections most frequent complication, significantly higher Dislodgements replacements also decreased markedly post-pandemic. Limited disrupted workflows likely contributed complication rates negatively impacted management patients, complications. decline highlights importance consistent care, infection prevention, remote monitoring, stronger resilience reduce risks future crises.

Pancreatic cancer (PC) is one of the most lethal malignancies worldwide and its incidence increasing. Chemotherapy often associated to limited efficacy, poor targeting systemic toxicity. In this work, hydrophilic gemcitabine (GEM), widely used in PC treatment alone or combination, was conjugated with vitamin E succinate (VES) encapsulated Soluplus® micelles. This prodrug approach facilitated encapsulation anticancer drug into self-assembled copolymer Soluplus®/VES-GEM micelles were optimized regarding ratio components preparation process. The small-sized (<80 nm), monodisperse, highly stable, efficiently retaining conjugate showing significant antiproliferative activity against BxPC3 cell line. To improve biofunctionalization properties prepared micelles, biomimetic modification membrane further attempted by co-extruding (BxPC3) nanovesicles Several protocols prepare BxPC3-modified outcomes analyzed detail. Overall, results pave way innovative PC-targeted nanotherapies maximizing GEM hydrophobic compartments high stability affinity. also highlight need higher resolution techniques characterize coating nanocarriers bearing shells.
