Ulcerative colitis (UC) is a chronic inflammatory bowel disease with rising incidence globally, whereas existing treatments exhibit significant limitations. Coix seed polysaccharide (CSP), component of traditional Chinese medicine known for its immunomodulatory and antioxidant properties, has not been thoroughly investigated role in UC. In this study, CSP was prepared via water extraction ethanol precipitation, protective effects mechanisms were evaluated using dextran sulfate sodium salt (DSS)-induced UC mouse model. The results demonstrated that significantly ameliorated DSS-induced symptoms, including weight loss, an elevated Disease Activity Index, colon shortening, increased levels cytokines, intestinal barrier damage. Moreover, reshaped the gut microbiota dysbiosis by increasing microbial diversity regulating abundance specific genera, such as Anaerotruncus. Metabolomic analysis revealed modulated 116 metabolites, particularly enhancing beneficial metabolite 3-hydroxybutyrate. Importantly, preventive effect on dependent could be transferred fecal transplantation. The cascade reaction of lipopolysaccharides (LPS), cell-free DNA (cfDNA), and reactive oxygen species (ROS), drives the development inflammatory bowel disease (IBD). Herein, we construct polyethylenimide (PEI)-L/D-tartaric acid (L/D-TA) complexes templated mesoporous organosilica nanoparticles (MON) (PEI-L/D-TA@MON) by mimicking biosilicification under ambient conditions within seconds. chiral nanomedicines include four functional moieties, wherein PEI electrostatically attracts cfDNA, tetrathulfide bonds reductively react with ROS, silanol groups adsorb LPS, L/D-TA enables recognition localization.
