Topical instilling of commercial artificial tears (cAT, containing 0.1 % hyaluronic acid) is widely employed to alleviate clinical manifestations mild dry eye disease (DED) by preventing the pathological change corneal epithelium. However, it showed limited therapy effectiveness on heavy DED which has further involved stroma, due its low stroma-available for acid (HA) resulting from barrier The present study developed a new microneedle-dosage form cAT (cAT-MN). Melanoma is a highly aggressive and metastatic malignancy, where current treatment methods often result in damage to healthy tissues, suboptimal therapeutic outcomes, immune-related side effects. Microneedles, as drug delivery system, offer advantages such localized administration, minimal invasiveness, high efficiency. In this study, we first synthesized tetradecyl-thiol-grafted PAMAM dendrimers, which significantly enhanced cellular uptake enabled sustained release of doxorubicin (DOX), improving cumulative Based on this, developed core-shell structured zwitterionic polymer-based microneedle system. The outer shell, loaded with the photothermal agent indocyanine green (ICG), achieved precise therapy under near-infrared irradiation, effectively targeting melanoma tissues. inner core, composed polymer matrix, encapsulated DOX-loaded enabling controlled prolonged through gradual swelling dendrimer expansion. Experiments show that system based grafted tetradecyl mercaptan has excellent anti protein adsorption properties, it can minimize cytotoxicity carrier improve efficiency delivery. This inhibited tumor growth synergistic photothermal-chemotherapy, reducing systemic toxicity bioavailability. platform provides promising strategy for targeted therapy, offering high-efficiency low-toxicity alternative.
