Herein, a rapid, clean, less expensive and environmentally friendly route to novel series of coumarins bearing thiazoles or 1,3,4-thiadiazoles was developed via grinding method under solvent conditions. Thus, 6-bromo-3-(2-bromoacetyl)-2-chromen-2-one treated with cyanothioacetamide produce thiazole-2-acetonitrile derivative 2, which then transformed iminocoumarins by reacting hydroxyaldehydes. Hydrolysis conc. HCl furnished coumarins. Furthermore, treatment compound 2 phenylisothiocyanate produced thioanilide interacts hydrazonoyl chloride and/or α-halocarbonyl compounds afforded 1,3,4-thiadiazole thiophene derivatives, consequently. All the newly prepared were screened against six pathogenic microorganisms. The results indicated that 7 12 most effective Bacillus pumilis, while 4b highly active Streptococcus faecalis. Also, 5b Enterobacter cloacae. Compounds higher compared reference drug ketoconazole fungi Saccharomyces cerevisiae.
