Introduction: Celastrol (Cel) is a widely used main component of Chinese herbal medicine with strong anti-inflammatory, antiviral and antitumor activities. In the present study, we aimed to elucidate cellular molecular protective mechanism Cel against diabetes-induced inflammation endothelial dysfunction. Methods: Type 2 diabetes (T2DM) was induced by db/db mice, osmotic pumps containing (100 μg/kg/day) were implanted intraperitoneally calibrated release drug for 28 days. addition, human umbilical vein cells (HUVECs) cultured in normal or high glucose palmitic acid-containing (HG + PA) media presence absence 48 h. Results: significantly ameliorated hyperglycemia-induced abnormalities nuclear factor (erythroid-derived 2)-like protein (Nrf2) pathway activity alleviated HG PA-induced oxidative damage. However, effect almost completely abolished HUVECs transfected short hairpin (sh)RNA targeting Nrf2, but not nonsense shRNA. Furthermore, PA reduced phosphorylation AMP-activated kinase (AMPK), autophagic degradation p62/Kelch-like ECH-associated 1 (Keap1), localization Nrf2. these catabolic pathways inhibited treatment HUVECs. compound C (AMPK inhibitors) AAV9-sh-Nrf2 Cel-induced Nrf2 activation angiogenesis mice. Discussion: Taking findings together, may be at least part attributed its effects reduce reactive oxygen species (ROS) through p62/Keap1-mediated activation.

Diabetic nephropathy (DN), one of the most common and severe microvascular complications diabetes, significantly increases risk renal failure cardiovascular events. A high-glucose environment can lead to mitochondrial dysfunction in macrophages, which, through remodeling energy metabolism, mediates polarization a pro-inflammatory phenotype contributes formation chronic inflammatory microenvironment. Recent studies have found that stimulation induces dysregulation nuclear factor erythroid 2-related 2 (NRF2) redox pathway leading generation oxidative stress (OS) further drives inflammation. Therefore, it is crucial fully understand how OS affects macrophage phenotypes functions following NRF2 inhibition. This review analyzes role induced by inflammation DN explores relationship between metabolism NAD⁺/NADH-SIRT3 axis, providing new therapeutic targets for targeting improve microenvironment vascular damage DN.

Diabetic peripheral neuropathy (DPN) is a vastly common and bothersome disorder with clinically challenging course of treatment affecting patients diabetes. This study aimed to evaluate the efficacy safety high dose oral N-acetyl cysteine (NAC) as adjuvant therapy on clinical outcome DPN. A prospective, randomized, parallel, open label, controlled trial. Ninety eligible DPN were randomly assigned either control group receiving standard care or NAC at 2400 mg/day for 12 weeks. Glutathione peroxidase (GPx), nuclear factor erythoid-2 related (NRF-2) tumor necrosis (TNF) measured baseline after weeks assess anti-oxidant anti-inflammatory properties. Michigan screening instrument (MNSI), Toronto score (TCNS), (DNS), Diabetes-39 quality life questionnaire (DQOL) pain assessed showed significant increase (p < 0.05) in NRF-2 by 25.3% GPx 100% decline 21.45% TNF-alpha levels versus controls that reported an TNF-alpha. HgbA1C AST significantly decreased (7.2 ± 1 vs 8 1.1, p = 0.028 29.1 55.4, 0.012) respectively. administration resulted MNSA, TCNS, DNS scores all scores. The QOL total anxiety energy mobility domain controls, 0.001. High administered modulated inflammation reducing increasing NRF2 controls. improved outcomes reflected improvement QOL. NCT04766450.

Atrazine (ATR), a widely used herbicide, poses significant environmental and health risks due to its high solubility adsorption in soil. ATR exposure can lead nephrotoxicity humans animals. Curcumin (Cur), an active compound Curcuma species, is renowned for antioxidant anti-inflammatory properties, with potential mitigate chronic disease risks. We hypothesized that the addition of Cur could alleviate renal impairment associated carried out experiments using mice as subjects. This study investigates whether attenuate ATR-induced by modulating mitophagy apoptotic pathways. Our findings illustrate consumption attenuates induced ATR, evidenced lowered serum concentrations uric acid (UA), blood urea nitrogen (BUN), creatinine (CRE), established biomarkers injury. Moreover, enhances defense mechanisms ATR-exposed mice, indicated elevated levels total capacity (T-AOC), catalase (CAT), glutathione peroxidase (GSH-Px), alongside reduced malondialdehyde (MDA). Histopathological electron microscopy analyses further corroborate these findings, showing organelle damage, particularly mitochondrial ridge breakage vacuolization, increased autophagic lysosomes. PINK1/Parkin-mediated autophagy, PINK1, Parkin, LC3BII, P62 compared ATR-treated mice. mitigates pathway, down-regulation apoptosis-related genes (Cytochrome C (Cyto-C), Caspase3, Caspase9) pro-apoptotic marker (Bax), along up-regulation anti-apoptotic (Bcl-2) at both transcriptional translational In summary, demonstrates nephroprotective properties against injury through enhancement autophagy display actions, underscoring curative potency treatment caused ATR.

Schisandra chinensis (S. chinensis) polysaccharide (SCP) is an active ingredient from S. used mainly for the treatment of diabetes, owing to its antioxidant, hypoglycemic, and lipidemic-modulating activities. A rat type II diabetes mellitus model was established by giving rats a high-fat diet streptozotocin (STZ) investigate protective effect SCP against renal injury in diabetic rats. It found this study that fasting blood glucose, serum lipids, creatinine, urea nitrogen levels were decreased, insulin sensitivity increased, pathological injuries kidney alleviated SCP-treated groups, indicating should have reduced C-reactive protein inhibited expression nuclear factors-κB related inflammatory factors tissue also regulated Nuclear factor (erythroid-derived 2) like-2, heme oxygenase-1, kelch-like ECH-associated protein-1, malondialdehyde content, increased superoxide dismutase activity. Furthermore, down-regulated fibronectin, α-SMA, transforming growth β1, p-Smad3, up-regulated Smad7 expression, mitigated collagen fiber deposition interstitium can be concluded mechanism alleviating may inhibiting inflammation, increasing antioxidant stress capacity, improving fibrosis

Macrophage-mediated inflammation plays essential roles in multiple-organ injury. Sinensetin (SNS) at least exhibits anti-inflammation, antioxidant, and antitumor properties. However, the underlying mechanism of SNS-targeted macrophage-mediated remains elusive. In present study, our results showed that SNS suppressed lipopolysaccharide (LPS)-induced to ameliorate lung liver injuries. Mechanistically, significantly inhibited M1-type macrophage polarization its NLRP3 inflammasome formation decrease tumor necrosis factor α (TNFα) IL-6 expression, while increasing IL-10 expression. Moreover, interacted activated SIRT1 promote NRF2 target gene SOD2 transcription, which subsequently decreased LPS-induced inflammation. knockdown impaired effects on inhibition polarization, formation, NRF2/SOD2 signaling. Taken together, is a potential promising natural active ingredient inflammatory injury via activating SIRT1/NRF2/SOD2
