Background: Microplastics (MPs) are small plastic fragments with diameters less than 5 mm in size and prevalent everyday essentials consumables. Large global production has now led to a flooding of MPs our natural environment. Due their detrimental impacts on the planet's ecosystems potentially health, have emerged as significant public health concern. In this pilot study, we hypothesize that exposure will negatively affect gut microbiota composition function, which metabolic reprogramming plays an important role. Methods: Using vitro experiments, three bacterial strains (Escherichia coli MG1655, Nissle 1917, Lactobacillus rhamnosus) were selected investigate exposure. The individually cultured anaerobic chamber exposed 1 µm polystyrene at various concentrations (0, 10, 20, 50, 100, 500 µg/mL) culture medium. Results: reduced growth all dose-dependent manner. Liquid chromatography mass spectrometry (LC-MS)-based untargeted metabolomics revealed differences multiple pathways, such sulfur metabolism amino sugar nucleotide metabolism. addition, extracted from C57BL/6 mice, 16S rRNA sequencing results showed upregulation Lactobacillales reduction Erysipelotrichales due Furthermore, targeted corroborated alterations microbial tryptophan energy producing glycolysis/gluconeogenesis pentose phosphate pathway. Conclusions: These findings provide evidence causes comprehensive changes healthy microbiota, may also insights into mechanistic effects humans.

Metal complexes are emerging as promising alternatives to traditional platinum-based cancer treatments, offering reduced side effects. However, understanding their cellular uptake and distribution quantifying presence at the single cell level remains challenging. Advanced imaging techniques, including transmission electron microscopy, synchrotron radiation X-ray fluorescence, energetic ion beam-based nuclear microscopy (scanning particle-induced emission, elastic backscattering spectrometry), allow detailed high-resolution visualization of structure morphology, high sensitivity for elemental detection with quantification within cells, construction 3D models metal distribution, positioning them powerful tools assessing compartmentalization complexes. The aim of the present study was to examine effects Astragalus polysaccharide (APS) on gut microbiota and serum metabolites in asthmatic mice. For this purpose, a total 36 BALB/c female mice were selected randomly classified into following groups: i) normal control group sensitized with phosphate‑buffered saline; ii) asthma challenged ovalbumin (OVA); iii) OVA + APS (2.5 g/kg) treatment group; iv) (5.0 v) (10 vi) dexamethasone (2 mg/kg) group, 6 each group. used establish mouse model asthma. In intragastrically administered at various doses 1 h prior stimulation. airway hyperreactivity (AHR) measured, hematoxylin eosin staining employed evaluate pulmonary inflammatory infiltration. addition, 16S rRNA sequencing ultra‑performance liquid chromatography‑tandem mass spectrometry detect changes metabolites. results revealed that compared improved inflammation eosinophil infiltration mice, microbial imbalance mainly manifested as low abundance Bacteroidetes high Firmicutes, yielding an increased F/B ratio. high‑dose Firmicutes reduced, increased, which thereby decreased ratio corrected imbalance. Through blood metabolomics, 145 105 significantly differential detected medium‑ groups, respectively. Moreover, Kyoto Encyclopedia Genes Genomes pathway enrichment analysis demonstrated metabolic pathways medium‑dose included biosynthesis unsaturated fatty acids arginine. Ergothioneine is a diet-derived micronutrient for humans. However, enzymes involved in the catabolism of ergothioneine human gut bacteria have not yet been identified. Herein, we characterize sulfidogenic pathway bacterial this micronutrient, which involves an unprecedented reductive desulfurization reaction catalyzed by members xanthine oxidoreductase family (XOR), class molybdenum-containing flavoproteins. Notably, first C-S bond cleavage known to be XORs. Evidence operation was gained through
