Intracranial aneurysms (IAs) are relatively common cerebrovascular anomalies. Melatonin could modulate inflammatory and offers neuroprotective effects, its role in IA has not been fully elucidated. An elastase-induced mouse model was constructed melatonin (150 mg/kg) administered to investigate therapeutic effects on IA. Aneurysm formation observed by bromophenol blue gelatin perfusion, the pathology changes mice were examined using hematoxylin-eosin (HE) staining. The potential mechanisms of treatment explored western blot, enzyme-linked immunosorbent, real-time qPCR, immunohistochemistry, flow cytometry. H2O2-reduced human brain vascular smooth muscle cells (HBVSMC) injury also constructed. circle Willis mice. alleviated thinning blood vessel walls disruption internal elastic lamina levels Bcl-2 significantly increased Bax cleaved caspase-3 decreased with treatment, suggesting reduced apoptosis. Furthermore, interleukin (IL)-1β, IL-6, tumor necrosis factor-alpha (TNF-α) HBVSMCs showed consistent results. krüppel-like factor 6 (KLF6) Importantly, regulatory T (Treg), hypoxia-inducible (HIF)-1α, nuclear 3-regulated (NFIL3), TCDD-inducible poly-ADP-ribose polymerase (TIPARP), monocytes mitigates modulating immune hypoxia-related factors. These findings provide an exploratory foundation for strategies

Given the growing challenges posed by an ageing population, particularly in Western countries, we aimed to investigate potential geroprotective effects of resveratrol and melatonin rats. The animals were treated with these two compounds starting at 3 months age continuing until 1 year or 2 years age. Using a multibiomarker approach, assessed DNA damage, telomere length, oxidative status their urine, liver, kidneys. Despite employing this experimental our results did not provide conclusive evidence across evaluated organs. However, observed sex-dependent differences response treatment. high potency compounds, further research is warranted explore incorporation into daily routines as strategy mitigate ageing-related effects.

Laboratory and animal studies indicate that melatonin exerts a negative impact on breast cancer progression metastasis. These actions are both receptor-dependent -independent. Of the two transmembrane receptors identified in humans, expresses only MT1. The aim of this study was to investigate expression MT1 hormone-receptor-positive, HER2-negative invasive ductal carcinoma postmenopausal women its possible correlations with clinicopathological parameters survival. A total 118 patients luminal A/B primary or without axillary metastases were identified. receptor immunohistochemically assessed as percentage stained cells weighted index (WI) (percentage multiplied by staining intensity). Most tumor samples (84.7%) metastasized lymph nodes (96%) positive for MT1, varying intensity. No statistically significant found between WI patient characteristics, nodes. survival analysis did not reveal effect risk recurrence

ABSTRACT The identification of protective agents for the treatment neurodegenerative diseases is mainstay therapeutic goal to modify disease course and arrest irreversible disability progression. Pharmacological therapies synergistically targeting multiple pathogenic pathways, including oxidative stress, mitochondrial dysfunction, inflammation, are prime candidates neuroprotection. Combination or synergistic therapy with melatonin, whose decline correlates altered sleep/wake cycle impaired glymphatic “waste clearance” system in diseases, has a great potential treat inflammatory states. Despite outcomes observed preclinical studies, mild poor were clinical settings, suggesting that melatonin combinations promoting actions at appropriate doses might be more suitable multifactorial disorders. In this review, we first summarize key pathways contributing cell protection its implication Alzheimer's (AD), Parkinson's (PD), sclerosis (MS). We remark major controversies field, mostly generated by lack common consensus optimal dosing, molecular targets, toxicity. Then, review literature investigating efficacy approved investigational neuroprotective melatonin‐containing hybrid molecules, both vitro animal models AD, PD, MS, as well add‐on settings. highlight rationale such focus on comparison single‐agent assays which an additive effect been achieved. conclude better characterization mechanisms underlying under neuroinflammation needs tackled advance successful translation combination melatonin‐based molecules.
