Abstract Background Clerodendrum speciosum is a hybrid of C. thomsonae and splendens . Many biological phytochemical studies have been performed on , but few conducted speciosum. Its methanol extract previously showed antioxidant activity in vivo via Caenorhabditis elegans model owing to its richness flavonoids phenylpropanoids. Results Characterization leaf volatile constituents (CSV) was gas chromatography linked mass spectrometry (GC–MS). This revealed the presence 29 metabolites that belonged mainly oxygenated monoterpenes, sesquiterpenes their compounds, fatty acids esters. The latter constituted predominant metabolites, whereas linoleic acid (30.64%) major metabolite. Liquid (LC–MS) ethyl acetate fraction leaves (CSE). resulted tentative assignment twenty-four peaks, six peaks were not identified. These identified phenylpropanoids, flavonoids, iridoid glycosides, phenolic derivatives also detected. notable antiarthritic as evidenced by significant inhibition albumin denaturation with an IC 50 32.50 μg/mL comparable diclofenac sodium (15.12 μg/mL). potent effect estimated 725.43 ± 5.95 mg TE/g, 333.82 19.9 2.1 0.08 mmol TE/g 3.69 0.3 EDTA/g. cupric-reducing capacity (CUPRAC), 2,2′-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS), phosphomolybdenum (PHD) metal-chelating (MCA) assays, respectively. It reasonable α -glucosidase -amylase 1.88 0.05 0.14 0.01 ACAE/g, ADME/TOPAKT assessment, processed prevalent components detected CSV, displayed acceptable pharmacodynamic, toxicity pharmacokinetic behaviors for most tested compounds except n -nonadecane -heptacosane -octacosane. Conclusion Thus, could serve promising treasure treatment many diseases such arthritis diabetes mellitus abundance terpenoidal compounds.
