Metabolic diseases have emerged as a leading cause of mortality from non-communicable diseases, posing significant global public health challenge. Although the association between ceramides (Cers) and metabolic is well-established, role acid sphingomyelinase (ASMase)/Cer pathway in these remains underexplored. This review synthesizes recent research on biological functions, regulatory mechanisms, targeted therapies related to ASMase/Cer conditions, including obesity, diabetes, non-alcoholic fatty liver disease, cardiovascular disease. The effects disease-related indicators, such glycolipid metabolism, insulin resistance, inflammation, mitochondrial homeostasis are elucidated. Moreover, this article discusses therapeutic strategies using inhibitors for inverse prevention treatment light possible efficacy blockade arresting progression diseases. These insights offered herein should provide insight into contribution offer tools develop interventions pathologies their severe complications.

The use probiotics, prebiotics synbiotics have been considered a potential promising strategy to regulate gut produce beneficial effects patients with conditions. For this reason, review aimed evaluate effectiveness prebiotics, symbiotics NAFLD NASH. Pubmed, Embase, Cochrane databases were consulted, PRISMA (Preferred Reporting Items Systematic Reviews Meta-Analysis) guidelines followed. clinical trials used study demonstrated that interventions could improve wide range markers inflammation, glycemia, insulin resistance, dyslipidemia, obesity, injury (decrease hepatic enzymes steatosis fibrosis). Although modulators do not play healing role, they work as an important adjunct therapy pathological processes involving its spectrums, either improving barrier or preventing formation toxic metabolites acting on system.

Gut dysbiosis, resulting from an imbalance in the gut microbiome, can induce excessive production of reactive oxygen species (ROS), leading to inflammation, DNA damage, activation immune system, and epigenetic alterations critical genes involved metabolic pathways. dysbiosis-induced inflammation also disrupt barrier integrity increase intestinal permeability, which allows gut-derived toxic products enter liver systemic circulation, further triggering oxidative stress, associated with diseases. However, specific metabolites, such as short-chain fatty acids (SCFAs), lactate, vitamins, modulate stress system through mechanisms, thereby improving function. microbiota diet-induced diseases, obesity, insulin resistance, dyslipidemia, hypertension, transfer next generation, involving mechanisms. Tryptophan (Trp) is an essential amino acid that can be metabolized via endogenous and exogenous pathways, including the Kynurenine Pathway, 5-Hydroxyindole Pathway (also Serotonin pathway), Microbial pathway. Of these, Trp metabolic pathways in gut have recently been extensively studied for their production of bioactive molecules. The microbiota plays important role host metabolism immunity, microbial metabolites influence development progression various diseases, inflammatory, cardiovascular neurological cancer, by mediating body’s immunity. This review briefly outlines crosstalk between microorganisms body, starting from three Trp. mechanisms which act on organism immunity are summarized, potential implications disease prevention treatment highlighted.

In recent years, although life expectancy has increased significantly, non-communicable diseases (NCDs) continue to pose a significant threat the health of global population. Therefore, eating habits have been recognized as key modifiable factors that influence people’s and well-being. For this reason, it is interesting study dietary patterns, since human diet complex mixture macronutrients, micronutrients, bioactive compounds, can modulate multiple physiological processes, including immune function, metabolism, inflammation. To ensure data we acquired were current relevant, searched primary secondary sources, scientific journals, bibliographic indexes, databases in last 15 years with most relevant articles. After search, observed all research on NCDs suggests critical factor shaping an individual’s outcomes. Thus, cardiovascular, metabolic, mental, dental, visual depends largely intake, nutritional behaviors.
