Abstract Background Rosmarinic acid (RA) has a wide range of beneficial effects on human health. On the other hand, RA been reported to induce metal-mediated reactive oxygen species (ROS) generation and DNA damage. However, its mechanism remains unknown. In this study, clarify underlying mechanism, we analyzed damage in isolated treated with analog isorinic acid. Results plus Cu(II), but not Fe(III), significantly increased 8-oxo-7,8-dihydro-2’-deoxyguanosine (8-oxodG) formation, an indicator oxidative damage, calf thymus DNA. Furthermore, comparison 8-oxodG formation induced by suggested that catechol groups could be associated their abilities form 8-oxodG. Interestingly, Cu(II) was markedly enhanced addition NADH, endogenous reductant. To elucidate Cu(II)-induced examined 32 P-labeled presence Cu(II). caused cleavage, which piperidine treatment, suggesting causes only strand breakage also base modification. inhibited catalase (H 2 O scavenger), bathocuproine (Cu(I) chelator), methional (scavenger variety ROS than • OH) typical OH scavengers SOD, indicating involvement H , Cu(I), OH. cleavage site analysis showing RA-induced site-specific (frequently at thymine some cytosine residues) supports OH, because without specificity. ABSTRACT Ocimum species have a great interest in different traditional medicinal systems. This study examined the chemical composition, antioxidant properties, enzyme inhibitory effects, and antibacterial antifungal activities of aerial parts gratissimum , americanum basilicum from Comoros Islands. The extracts were analyzed using high‐performance liquid chromatography‐mass spectrometry (HPLC‐MS) to determine their composition. Antioxidant activity was assessed 2,2‐Diphenyl‐1‐picrylhydrazyl (DPPH), 2,2′‐Azino‐bis(3‐ethylbenzothiazoline‐6‐sulfonic acid) diammonium salt (ABTS), cupric reducing capacity (CUPRAC), ferric power (FRAP), chelating ability, phosphomolybdenum radical scavenging assays. Enzyme against acetylcholinesterase (AChE), butrylcholinesterase (BChE), tyrosinase, amylase, glucosidase evaluated spectrophotometric methods. Antibacterial tested broth microdilution method selected pathogenic microorganisms. enzymes proteins silico methods with biomolecules these plants. In addition, 111 metabolites identified advanced HPLC/MS techniques. most significant number detected compounds derivatives hydroxycinnamic acids, followed by flavonoid glycosides aglycones hydroxybenzoic acids. All three exhibited activities, O. best‐reducing abilities CUPRAC FRAP assays revealed that had potent effect on tyrosinase (48.01 ± 3.89 mg kojic acid equivalent [KAE]/g), amylase (1.08 0.02 mmol acarbose [ACAE]/g). tests demonstrated possess broad‐spectrum activity. Molecular docking results showed remarkable binding energies target proteins. molecular dynamics simulations chicoric MurE Staphylococcus aureus complex as promising drug candidate. These findings support medical nutraceutical uses suggest possibilities for natural functional applications.

Abstract Introduction Skin cancer poses a significant health risk globally, necessitating effective and safe therapeutic interventions. Epigallocatechin‐3‐gallate (EGCG) from green tea rosmarinic acid (RA) herbs like rosemary offer promising anticancer properties. Combining these compounds may enhance their effectiveness, prompting the need for reliable analytical method to quantify them. Objective Herein, we present development validation of high‐performance thin‐layer chromatography (HPTLC) concurrent quantification EGCG RA in lipid‐based nanoparticles biological samples. Methodology The underwent optimisation through design experiments (DoE), resulting establishment robust chromatographic conditions. separation process utilised aluminium HPTLC plates coated with silica gel 60 F 254 as stationary phase, mobile phase comprising ethyl acetate, toluene, formic acid, methanol ratio 4:4:1:1 v/v. Results retention factor ( R f ) values obtained were 0.38 0.61 RA. demonstrated linearity over range 100–500 ng/band both excellent correlation coefficients. Limits detection determined, indicating high sensitivity. Precision evaluations revealed relative standard deviation below 2%, ensuring reproducibility. Recovery assays nanoparticles, plasma, urine samples recoveries (96.2%–102.1%). Forced degradation studies minimal under various stress conditions, photolytic showing least impact. Conclusion developed offers rapid, sensitive, approach quantifying RA, laying groundwork further investigation agents alone combination therapies.

Rosmarinus officinalis L. compounds, especially its main polyphenolic carnosic acid (CA) and rosmarinic (RA), influence various facets of cancer biology, making them valuable assets in the ongoing fight against cancer. These two secondary metabolites exhibit formidable antioxidant properties that are a pivotal contributor development Their antitumor effect has been related to diverse mechanisms. In case CA, it capacity induce cell death cells through rise ROS levels within cells, inhibition protein kinase AKT, activation autophagy-related genes (ATG) disrupt mitochondrial membrane potential. Regarding RA, actions encompass apoptosis induction caspase activation, proliferation by interrupting cycle progression epigenetic regulation, antioxidative stress-induced DNA damage, interference with angiogenesis curtail tumor growth. To understand molecular interaction between rosemary compounds (CA RA) is involved inflammation, S100A8, we have performed series docking analyses using available three-dimensional structures (PDBID: 1IRJ, 1MR8, 4GGF). The ligands showed different binding intensities active sites target molecules, except for CA 1MR8 protein.

Rosmarinic acid (RosA) is a natural polyphenol compound that has been shown to be effective in the treatment of inflammatory disease and variety malignant tumors. However, its specific mechanism for lung adenocarcinoma (LUAD) not fully elucidated. Therefore, this study aims clarify RosA LUAD by integrating bioinformatics, network pharmacology vivo experiments, explore potential active ingredients traditional Chinese medicine treating LUAD. Firstly, was used screen targets, LUAD-related differential expressed genes (DEGs) were acquired from GEO database. The intersection regulated (RDEGs) obtained through Venn diagram. Secondly, GO KEGG enrichment analysis RDEGs performed, protein–protein interaction networks (PPIs) constructed identify visualize hub RDEGs. Then, molecular docking between further evaluation carried out using bioinformatics predictive value Finally, verified establishing xenograft model NSCLC nude mouse. Bioinformatics other showed that, compared with control group, expressions MMP-1, MMP-9, IGFBP3 PLAU tissues significantly up-regulated, PPARG FABP4 down-regulated, these had In experimental results could inhibit growth transplanted tumors mice bearing cancer cells, reduce positive expression Ki67 tumor tissue, hinder proliferation cells. Upregulated activating PPAR signaling pathway increases level ROS promotes apoptosis addition, can also MMP-9 IGFBP3, migration invasion tissue This demonstrated induce regulating proliferation, thereby exerting anti-LUAD effects. provides new insight into therapeutic avenue
