Abstract Background and aim Ulcerative colitis (UC) is characterized by complex immunological interactions involving CD4 T cell subsets the NLRP3 inflammasome, which influence inflammatory responses. This investigation focused on delineating activation profiles of these components their correlation with disease severity activity, assessing diagnostic implications in UC. Methods We conducted immunohistochemistry ELISA assays to measure markers expression inflammasome UC patients versus controls. Findings were validated using analysis, molecular docking ROC curves. Results displayed increased Th1 (T-bet, TNF-α), Th2 (GATA3, IL-6), Th17 (RORγt, IL-17, IL-22, IL-23) Additionally, cytokines (IL-2 IL-4) significantly elevated severe UC, while Treg (FOXP3, IL-10, TGF-β1) only mild-to-moderate Enhanced activation, indicated NLRP3, Caspase-1, IL-1β levels. These patterns, confirmed through analysis docking, underscored strong correlations among T-bet, GATA3, supporting proposed NLRP3/T-bet/GATA3 axis. axis, along other biomarkers, showed associations severity, Mayo score, UCEIS, demonstrated relatively high value. Conclusion The axis provides a referable strategy for multi-targeted combined treatment may serve as potential biomarkers enhancing accuracy guiding therapy.

Ulcerative colitis (UC) is a chronic idiopathic inflammatory disease affecting the colon and rectum. Characterized by recurrent attacks, UC often resistant to traditional anti-inflammatory therapies, imposing significant physiological, psychological, economic burdens on patients. In light of these challenges, innovative targeted therapies have become new expectation for patients with UC. A crucial pathological feature impairment intestinal mucosal barrier, which underlies aberrant immune responses inflammation. Intestinal stem cells (ISCs), differentiate into epithelial cells, play central role in maintaining this barrier. Growing studies proved that regulating regeneration differentiation ISC promising approach treating Despite progress, there dearth comprehensive articles describing ISCs This review focuses importance barrier discusses latest findings functions, markers, their regulatory mechanisms. Key pathways involved regulation, including Wnt, Notch, Hedgehog (HH), Hippo/Yap, autophagy pathways, are explored detail. Additionally, examines recent advances ISC-targeted UC, such as natural or synthetic compounds, microbial preparations, Chinese medicine (TCM) extracts transplantation therapy. aims offer novel therapeutic insights strategies who long struggled

Abstract The success of graphene oxides has gained extensive research interests in developing novel 2D nanomaterials (NMs). WS 2 nanosheets (NSs) are transition metal‐based NMs, but their toxicity is unclear. In this study, we investigated the oral NSs to mouse intestines. Male mice were administrated with vehicles, 1, 10, or 100 mg/kg via intragastric route, once a day, for 5 days. results indicate that did not induce pathological ultrastructural changes There minimal trace elements exposure W accumulation, and only Co levels dose‐dependently increased. Lipid droplets observed all groups mice, lipidomics data significantly decreased four lipid species, belonging phosphatidylcholine (PC). proteins regulating autophagic lipolysis, namely, LC3, lysosomal associated membrane protein (LAMP2) perilipin (PLIN2), increased, it was statistically different LC3. study suggest repeated induced influences on injury, element balance, autophagy, profiles intestines, indicating relatively high biocompatibility intestine route.

Abstract With the frequent occurrence of heatwaves, heatstroke (HS) is expected to become one main causes global death. Being a multi-organized disease, HS can result in circulatory disturbance and systemic inflammatory response, with gastrointestinal tract being primary organs affected. Intestinal damage plays an initiating promoting role HS. Multiple pathways integrity intestinal epithelial barrier due heat stress hypoxia brought on by blood distribution. This usually leads leakage as well infiltration metastasis toxins pathogenic bacteria cavity, which will eventually cause inflammation whole body. A large number studies have shown that after involves body’s disruption oxidative balance, disorder tight junction proteins, massive cell death, microbial imbalance. Based these mechanisms, protecting regulating immune responses are effective treatment strategies. To better understand pathophysiology this complex process, review aims outline potential processes possible therapeutic strategies for recent years.

Abstract The recurrent pathological inflammation of the gut is a major concern in diabetic patients. With failure anti-inflammatory or drugs to limit relapse colon demands unearthing mechanistic details underlying higher incidences colitis Here we report enrichment DPP4 livers and blood samples humans mice models diabesity that parallel development colitis. Overexpression exacerbates hepatic silencing impairs experimental induced by DSS STM. Mechanistically, identified liver attenuates gut-autophagic response trigger enteric cell apoptosis, reduced mucin secretion, compromised barrier leading high infiltration immune cells secreting inflammatory cytokines establishing inflammation. Thus, liver-DPP4-mediated autophagy inhibition key pathway diabesitic
