Breast cancer represents one of the most prevalent malignant neoplasms affecting women, and its pathogenesis has garnered significant scholarly interest. Research indicates that progression breast is intricately regulated by glucose metabolism. Under hypoxic conditions within tumor microenvironment, cells generate ATP essential biosynthetic precursors for growth via glycolytic pathway. Notably, phosphoglycerate kinase 1 (PGK1) intimately associated with regulation hypoxia-inducible factors in plays a crucial role modulating processes. Further investigation into PGK1 anticipated to identify novel therapeutic targets strategies. This review consolidates current research on metabolism function conditions. It aims offer theoretical foundation elucidating mechanisms underlying metastasis, thereby facilitating development innovative treatment approaches.

Breast cancer is one of the most common female malignant tumors, which seriously endangers human health. Glucose metabolic reprogramming in rapidly proliferating cancers drives increased glycolysis to meet energy needs, promoting tumor growth, acidifying microenvironment, and impairing immune function, diminishes therapeutic efficacy. Circular RNAs (circRNAs), as key regulators cellular processes, are increasingly recognized for their involvement cancer. Concurrently, specific circRNAs could be released by cells via exosomes facilitate intercellular communication, significantly impacting glucose metabolism, progression, therapy resistance. However, role breast mechanisms regulating metabolism remain unclear. Therefore, elucidating these regulatory pathways provide valuable insights developing targeted strategies exploit vulnerabilities improve prognosis

Dietary behaviors significantly influence tumor progression, with increasing focus on high-salt diets (HSD) in recent years. Traditionally, HSD has been regarded as a major risk factor for multiple health issues, including hypertension, cardiovascular disease, kidney cancer, and osteoporosis. However, studies have uncovered novel aspect of HSD, suggesting that may inhibit growth specific pathological conditions by modulating the activity immune cells infiltrate tumors enhancing effectiveness PD-1 immunotherapy. This review focused duel molecular mechanisms cancer development, which are based microenvironment, gut microbiota, involvement sodium transporter channels. The objective this is to explore whether could be potential future oncological therapeutic strategy under situation.

Abstract Background Myeloid-derived suppressor cells (MDSCs) in tumor microenvironment reduce the efficacy of immunotherapy. PKN2 plays a role colon cancer, but its function esophageal cancer (EC) remains unclear. This study investigated expression MDSCs derived from EC tissues and determined whether regulates immunosuppressive activity by mediating fatty acid oxidation (FAO). Materials methods was GEO database, patients, 4-NQO-induced mice, as well different types immune cells. The effect on polymorphonuclear myeloid-derived (PMN-MDSCs) co-culture PMN-MDSCs CD4 + /CD8 T patient-derived organoids autologous performed to observe PMN-MDSCs. Results is highly expressed compared normal tissues, especially tumor-infiltrated Overexpressing contributes vitro. PKN2-overexpressing inhibited killing ability cytotoxic lymphocytes promoted organoid growth. promotes FAO via CPT1B (a key enzyme FAO). Mechanistically, transcription upregulating STAT3 phosphorylation. Conclusions increased human mouse tissues. enhancing facilitating phosphorylation transcription, which turn leads CPT1B-mediated Targeted inhibition expected improve immunotherapeutic patients.
