Sulfonamide antibiotics (SAs) are widely used in medicine, animal husbandry and aquaculture, excessive intake of SAs may pose potential toxicity to organisms. The toxicological mechanisms two classical SAs, sulfamerazine (SMR) sulfamethoxazole (SMT), were investigated by molecular docking, DFT multi-spectroscopic techniques using HSA BSA as model proteins. quenching HSA/BSA endogenous fluorescence SMR was higher than that SMT due the stronger binding effect pyrimidine ring on compared oxazole ring, result consistent with predicted calculations. Thermodynamic parameters show is an exothermic process proceeds spontaneously (ΔG < 0). Marker competition experiments illustrate site SMR/SMT serum albumin located subdomain IIIA. combination mainly realized hydrogen bond hydrophobic interaction, concept also supported modeling. reduced α-helix content induced indicates a greater stretching protein structure SMR/SMT-HSA/BSA. results could provide useful information hazards response growing concern toxic threat
