The GRAS (generally recognized as safe) strain Pediococcus acidilactici is well known for its antibacterial and probiotic functions. Furthermore, P. has excellent high temperature salt resistance, it an ideal host the production of food enzymes, additives, pharmaceuticals. In this regard, desirable feasible to enhance these products through metabolic engineering acidilactici. However, rare gene expression elements greatly obstruct development study, we screened constructed a library promoter-5'-UTR (PUTR) complexes in DY15 regulating at transcription translation levels. post-log phase, mRNA protein level ranges 90 native PUTRs were 0.059-2010% 0.77-245%, respectively, P32 promoter. Besides, several exhibited great stability under temperature, salt, ethanol stress. We analyzed structure obtained conserved regions promoter 5'-UTR. Based on identified core PUTRs, panel combinatorial with higher stable strongest PUTR was 853% level. Finally, applied optimize aminotransferase improve phenyllactic acid (PLA) DY15. achieved yield 950.6 mg/L, which 79.2% than wild-type strain. These results indicated that gradient strength had potential precisely achieve various goals
