Objectives: This study investigates the effectiveness of an herbal formulation, STRESSLESS (ST-65), which combines ashwagandha (Withania somnifera) and bacopa (Bacopa monnieri), on SH-SY5Y human neuroblastoma cells. Given rising interest in natural compounds for neuroprotection stress alleviation, we aimed to explore cellular molecular effects this formulation. Methods: Utilizing a nuclear magnetic resonance (NMR) metabolomic approach ultra-high-performance liquid chromatography-high-resolution mass spectrometry (UHPLC-HRMS), identified key bioactive ST-65, including withanolides from bacosides bacopa. Results: Our findings indicate that ST-65 treatment significantly alters metabolic profile Key changes included increased levels metabolites linked neuroprotection, energy metabolism, antioxidant defense. Abstract Background Huntington's disease (HD) could cause progressive motor deficits, psychiatric symptoms, and cognitive impairment. With the increasing use of pharmacotherapies theoretically target neurotransmitters, incidence HD is still not decreasing. However, molecular pathogenesis have been illuminate. It momentous to further examine HD. Methods The next generation sequencing dataset GSE105041 was downloaded from Gene Expression Omnibus (GEO) database. Using DESeq2 in R bioconductor package screen differentially expressed genes (DEGs) between samples normal control samples. ontology (GO) term REACTOME pathway enrichment were performed on DEGs. Meanwhile, using Integrated Interactions Database (IID) database Cytoscape software construct protein–protein interaction (PPI) network module analysis, identify hub with highest value node degree, betweenness, stress closeness scores. miRNA-hub gene regulatory TF-hub constructed analyzed. Receiver operating characteristic curves analysis for diagnostic genes. Results We identified 958 DEGs, consisting 479 up regulated DEGs down GO terms analyses by g:Profiler online results revealed that mainly enriched multicellular organismal process, developmental signaling GPCR MHC class II antigen presentation. Selye described stress as a unified neurohormonal mechanism maintaining homeostasis. Acute system activation is adaptive through neurocognitive, catecholaminergic, and immunomodulation mechanisms, followed by reset via cortisol. Stress components, the sympathoadrenomedullary system, hypothalamic-pituitary-adrenal axis, limbic structures are implicated in many chronic diseases establishing an altered homeostatic state, allostasis. Consequent "primary disorders" were popularly accepted, with phenotypes based on conditions such Cushing syndrome, pheochromocytoma, adrenal insufficiency. Cardiometabolic major depressive disorders candidates for hypercortisolemic etiology, contrasting "hypocortisolemic symptom triad" of sensitivity, fatigue, pain. However, acceptance etiology requires cause-and-effect associations, practical utility therapeutics altering function. Inherent predispositions to perturbations may be relevant. Background The hypothalamic-pituitary-adrenal (HPA) axis and the hypothalamic-pituitary-thyroid (HPT) are two neuroendocrine systems that play a crucial role in maintaining homeostasis regulating mood alterations.
