Pancreatic β-cell damage is a critical pathological mechanism in the progression of obese type 2 diabetes mellitus (T2DM). However, exact underlying remains unclear. We established an T2DM mouse model via high-fat diet feeding. The protein expression profiles pancreatic tissues from normal and mice were analyzed, revealing that nuclear factor activated T cells 5 (NFAT5) ferroptosis are potential mediators mechanisms mice. In vitro, high glucose palmitate treatment resulted increased NFAT5 translocation MIN6 cells. Inhibition by shRNA significantly reduced improved reduction insulin secretion palmitic acid (PG)-treated Luciferase reporter chromatin immunoprecipitation (ChIP) assays confirmed ability to bind peroxiredoxin (PRDX2) promoter, leading downregulation PRDX2 transcription. Subsequent rescue experiments involved PG-induced inhibiting PRDX2.
