Exploring cell death pathways in oral cancer: mechanisms, therapeutic strategies, and future perspectives

Oral
squamous
cell
carcinoma
(OSCC)
represents
a
significant
global
health
challenge,
characterized
by
aggressive
progression
and
poor
therapeutic
response
despite
advances
in
treatment
modalities.
This
review
provides
comprehensive
analysis
of
diverse
death
mechanisms
OSCC,
encompassing
traditional
pathways
(apoptosis,
autophagy,
necrosis),
newly
(ferroptosis,
pyroptosis,
necroptosis),
emerging
(cuproptosis,
anoikis,
parthanatos,
entosis).
By
examining
the
molecular
basis
these
pathways,
particularly
crucial
roles
p53
signaling
miRNA
regulation,
we
highlight
how
their
dysregulation
contributes
to
resistance
tumor
progression.
The
synthesizes
recent
evidence
demonstrating
complex
interplay
between
ten
distinct
impact
on
microenvironment
immune
response.
We
evaluate
innovative
approaches
that
target
including
novel
small
molecules,
combination
strategies,
immunomodulatory
treatments
exploit
specific
enhance
efficacy.
Special
attention
is
given
personalized
medicine
strategies
consider
individual
characteristics
pathway
profiles.

Pancreatic
cancer,

prognosis,
exhibits
a
marked
resistance
to
conventional
chemotherapy
and
immunotherapy,

study
is
designed
assess
potential
antineoplastic
efficacy
combined
regimen
involving
tetrandrine,
plant-derived
alkaloid,
autophagy
inhibitors
in
context
pancreatic
cancer.
Electron
microscopy
immunoblots
showed
that
tetrandrine
promoted
formation
autophagosomes
upregulation
LC3II
downregulation
p62
expression,
indicating
induced
cancer
cells.
Western
blot
revealed
inhibited
phosphorylation
AKT
mTOR,
as
well
expression
Bcl-2,
while
upregulating
Beclin-1
expression.
Moreover,
transcription
protein
ATG7.
Following
combination
apoptotic
rate
cell
death
significantly
increased

cancer.
The
transitional
cell
carcinoma
histotype
aggressive
and

therapies
are
ineffective.
We
investigated
anti-proliferative
effect
of
quercetin,
natural
flavonoid,
in
combination
with
alkylating
agent
mafosfamide
(MFA)
on
two
human
bladder
cancer
lines,
namely
RT112
J82,
representing
progression
from
low-grade
to
high-grade
tumors,
respectively.
In
both
types,
combined
treatment
led
synergic
reduction
viability
confirmed
by
index
less
than
one,
though
different
biological
responses
were
noted.
J82
cells,
MFA
alone
and,
lesser
extent,
quercetin
caused
cycle
arrest
G2/M
phase,
but
only
triggered
apoptotic
death.

autophagy,
evidenced
autophagosome
formation
increase
LC-3
lipidation.
Interestingly,
synergistic

observed
when
cells
pre-treated
for
24
h
before

recent
years,
the
role
of
gut
microbiota
(GM)
in
bladder
cancer
has
attracted
significant
attention.
Research
indicates
that
GM
not
only
contributes
to
carcinogenesis
but
also
influences
efficacy
adjuvant
therapies
for
cancer.
Despite
this,
interventions
targeting
have
been
widely
employed
prevention
and
treatment
cancer,
mainly
due
incomplete
understanding
complex
interactions
between
host
flora.
Simultaneously,
aberrantly
expressed
non-coding
RNAs
(ncRNAs)
frequently
associated
with
playing
crucial
roles

as

proliferation,
