In multiple areas such as science, technology, and economic activities, it is necessary to unify the management of repetitive tasks or concepts by standardization obtain best order high efficiency. Organoids, living tissue models, have rapidly developed in past decade. Organoids can be used repetitively for vitro culture, cryopreservation, recovery further utilization. Because organoids recapitulate parental tissues' morphological phenotypes, cell functions, biological behaviors, genomic profiles, they are known renewable "living biobanks". cover two mainstream fields: Adult stem cell-derived (also patient-derived organoids) induced pluripotent and/or embryonic organoids. Given increasing importance development new drugs, standardized operation, all steps organoid construction an important guarantee ensure quality products. this review, we systematically introduce operation procedures, laboratory construction, available documents related culture that been published so far. We also proposed challenges prospects field.

Abstract Lung adenocarcinoma (LUAD) exhibits differences between the sexes in incidence, prognosis, and therapy, suggesting underexplored molecular mechanisms. We conducted an integrative multi-omics analysis using Clinical Proteomic Tumor Analysis Consortium (CPTAC) The Cancer Genome Atlas (TCGA) datasets to contrast transcriptomes proteomes sexes. used TIGER analyze TCGA-LUAD expression data found sex-biased activity of transcription factors (TFs); we PTM-SEA with CPTAC-LUAD proteomics kinase activity. combined these construct a kinase-TF signaling network discovered druggable pathways linked cancer-related processes. also significant sex biases clinically relevant TFs kinases, including NR3C1, AR, AURKA. Using PRISM drug screening database, identified potential sex-specific drugs, such as glucocorticoid receptor agonists aurora inhibitors. Abstract Osteosarcoma (OS) exhibit intra- and inter-heterogeneity, complicating the exploration of effective therapeutic strategies. Traditional in vitro vivo models are limited inheriting biological genomic heterogeneities OS patients, even features on tumor microenvironment. The prolonged generation time current makes drug development slow is not suitable to clinically rapid timing. Here, we introduce methods for generating biobanking patient/PDX-derived osteosarcoma organoids (OS PD(X)Os) that recapitulate histological, their paired patients. PD(X)Os can be generated quickly with high reliability or transplanted immunodeficient mice. We further demonstrate an immune-featured PD(X)O (named iOS) model its method testing personalized chemotherapy response, immune strategy target development, such as a novel PRMT5 MTA inhibitor ARPN2169 MTAP-deleted OS. Our studies show iOS maintain many typical could rapidly employed investigate patient-specific Additionally, our biobank establishes rich resource basic, translational clinical researches.

This study explored the comorbid mechanisms between Crohn's disease (CD) and chronic obstructive pulmonary (COPD) using bioinformatics analysis. From Gene Expression Omnibus (GEO) microarray dataset, 349 common differentially expressed genes (coDEGs) were identified, 8 shared hub found: CCL2, CXCL1, TLR2, ICAM1, PTPRC, ITGAX, PTGS2, MMP9, which vital for immune function regulation of inflammatory responses. In addition, also analyzed association coDEGs cell infiltration single-sample gene set enrichment algorithm (ssGSEA). Potential drugs related to these identified connectivity map (CMap). These findings provided new perspectives understanding interaction CD COPD.
