Background/Objectives: Atopic dermatitis (AD) is a chronic inflammatory skin disorder that has attracted global attention, and alkaloids from Plumula Nelumbinis have been shown to anti-inflammatory activity. Fermentation used for the structural modification of natural compounds improve bioavailability activity, but AD therapeutic efficacy mechanism fermented (FPN) are still unclear. Methods: The potential targets FPN were preliminarily screened using network pharmacology, then PCR WB prove effect in AD. Results: Network pharmacology indicated mTOR Jun key experiments vitro showed could effectively block AKT/mTOR AKT/Jun-mediated signaling pathways. Moreover, can also alleviate SDS-induced inflammation zebrafish. It found activity was enhanced by Aspergillus oryzae fermentation, oil phase fermentation product better which may be due microbial changing structure original alkaloids. Conclusions: This study elucidated mechanisms derived against AD; it provide scientific basis development new drugs

With the increasing attention of modern medicine to natural medicinal agents, Inonotus obliquus (chaga), a macrofungus with remarkable value, has gradually garnered widespread academic interest. This paper reviews primary bioactive components I. in recent years, including polysaccharides, phenolic compounds, and triterpenoids, which exhibit diverse pharmacological effects such as antioxidant, anti-inflammatory, immunomodulatory, antitumor activities. It further discusses how these enhance human health disease resistance through distinct biological mechanisms, activation antioxidant systems, regulation immune responses, modulation apoptosis pathways. Additionally, article explores biosynthetic pathways metabolites their relevance. Finally, we summarize potential resource envisage its future applications clinical drug development. review aims provide novel perspectives for cultivation, utilization, industrial-scale exploitation obliquus.
