Abstract Cannabidiol (CBD) is a non-intoxicating cannabinoid from cannabis sativa that has demonstrated efficacious against inflammation, which can be considered as potential drug for arthritis treatment. However, the poor solubility and low bioavailability limit its clinical application. Here, we report an effective strategy to fabricate CBD-loaded poly lactic-co-glycolic acid nanoparticles (CBD-PLGA-NPs). The CBD-PLGA-NPs exhibited spherical morphology average diameter of 238 nm. CBD was sustained release CBD-PLGA-NPs, improved CBD. Primary chondrocytes rat pups were isolated, LPS used induce inflammation in vitro simulate osteoarthritis (OA). effectively protect damage cell viability. What’s more, according results CCK-8 assay, hematoxylin-eosin staining, safranin O immunofluorescence real-time polymerase chain reaction observed significantly suppressed LPS-induced primary chondrocyte expression inflammatory cytokines, including interleukin 1β (IL-1β), 6 (IL-6), tumor necrosis factor-α (TNF-α) matrix metalloproteinase 13 (MMP-13). Remarkably, also showed better therapeutic effects inhibiting degradation extracellular than equivalent solution. In general, fabrication good protection promising system Significance study order improve CBD, described development simple efficient (CBD-PLGA-NPs) treating damaged. fabricated could enhance chondroprotective by factors, increasing cellularity, improving structural changes, regarded treat OA.

Cannabidiol (CBD) is the primary non-psychoactive chemical from Cannabis Sativa, a plant used for centuries both recreational and medicinal purposes. CBD lacks psychotropic effects of Δ9-tetrahydrocannabinol (Δ9-THC) has shown great therapeutic potential. exerts wide spectrum at molecular, cellular, organ level, affecting inflammation, oxidative damage, cell survival, pain, vasodilation, excitability, among others, modifying many physiological pathophysiological processes. There evidence that may be effective in treating several human disorders, like anxiety, chronic psychiatric pathologies, cardiovascular diseases, even cancer. Multiple cellular pre-clinical studies using animal models disease trials have an overall safe profile. In this review article, we summarize pharmacokinetics data, putative mechanisms action CBD, reported to give comprehensive list findings major attributed compound.

This study aimed to investigate the anti-inflammatory effect of walnut-derived peptide LPLLR (LP-5) in oxidative stress-induced Caco-2 cells and model mice. The results showed that LP-5 significantly increased activities CAT SOD, while decreased levels MDA both vitro vivo. stress signaling was regulated by promoting Nrf2 nuclear transfer, up-regulating HO-1 expression, down-regulating Keap1 expression. As demonstrated molecular docking, could effectively bind Keap1. Additionally, inhibited activation NLRP3 inflammasome via expression NLRP3, ASC, caspase-1, reducing release inflammatory cytokines, including IL-18, IL-1β, TNF-α, thereby preventing cell apoptosis. Moreover, production IL-10 iNOS COX-2 cells. These data suggested ameliorated stress-mediated inflammation inhibiting activating Nrf2/Keap1 signaling.

Suk-SaiYasna (SSY) is a well-documented traditional Thai herbal formula in the Royal Scripture of King Narai’s Traditional Medicine. SSY contains Cannabis sativa leaves as key ingredient and has traditionally been used to promote sleep, alleviate stress-related symptoms, stimulate appetite. This study aimed investigate neuroprotective effects mouse model unpredictable chronic mild stress (UCMS)-induced cognitive impairment explore underlying mechanisms, particularly antioxidant enzyme pathways. Behavioral tests, including Y-maze test, novel object recognition Morris water maze demonstrated that UCMS-exposed mice exhibited compared non-stress mice. However, treatment significantly improved learning memory performance Mechanistic studies revealed reduced lipid peroxidation hippocampus frontal cortex, brain regions affected by stress. Furthermore, UCMS activity enzymes superoxide dismutase (SOD) catalase (CAT), whereas restored their activity, indicating antioxidative vivo. Gene expression analysis further regulates oxidative via Nrf2/Keap1 signaling pathway. In vitro using 2,2′-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid (ABTS) 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay confirmed radical scavenging activities its components, demonstrating significant potential. Phytochemical identified delta-9-tetrahydrocannabinol, delta-9-tetrahydrocannabinolic A, cannabinoids bioactive compounds SSY, along with potent antioxidants such gallic acid, myricetin, myristicin, piperine, costunolide, gingerol. These findings suggest mitigates UCMS-induced function through properties multiple pathways, activities, modulating Nrf2-Keap1 pathway, inducing HO-1, NQO1 mRNAs, other enzymes. work bridges medicine modern neuropharmacology.
