Cannabinoids include both endogenous endocannabinoids and exogenous phytocannabinoids, such as cannabidiol (CBD), have potential therapeutic agents in cancer treatment due to their selective anticancer activities. CBD exhibits antioxidant pro-oxidant effects depending on its concentration cell types. These properties allow influence oxidative stress responses potentially enhance the efficacy of antitumor therapies. In this study, we treated U87MG glioma cells with low dose (1 μM) evaluated molecular effects. Our findings indicate that reduced viability by 20% (p < 0.05) through alteration mitochondrial membrane potential. The redox status caused an attempt rescue functionality nuclear localization GABP transcription factor involved mitochondria biogenesis. Moreover, increase autophagic flux, supported Beclin-1 ratio LC3-II/LC3-I. Due alteration, pro-apoptotic proteins were induced without activating apoptotic effectors Caspase-3 or Caspase-7. study NRF2 ubiquitin-binding protein p62 expression revealed levels CBD-treated cells. conclusion, low-dose makes more vulnerable cytotoxic effects, reducing dynamics while increasing flux systems. This explains mechanisms which respond treatment. highlight CBD, suggesting modulating autophagy pathways could represent a promising strategy for glioblastoma

ABSTRACT Ferroptosis plays a crucial role in the pathogenesis of osteoarthritis (OA), and inhibition chondrocyte ferroptosis effectively alleviates OA progression. Krüppel‐like factor 9 (KLF9) is demonstrated to be upregulated OA, but its molecular mechanism remains unclear. The study aimed investigate KLF9 Primary chondrocytes were treated with IL‐1β establish an cell model, showed that was highly expressed IL‐1β‐incubated chondrocytes. Knockdown alleviated IL‐1β‐induced degeneration. In addition, decreased methylation proportion gene promoter. DNA methyltransferase 1 (DNMT1) directly bound promoter, overexpression DNMT1 inhibited expression by promoting promoter Subsequently, shRNA pcDNA‐CYP1B1 individually or altogether transfected into Cytochrome P450 1B1 (CYP1B1) chondrocytes, abrogated inhibitory effect on ferroptosis. Moreover, Ferrostatin‐1 (Fer‐1, inhibitor ferroptosis) reversed promotion Finally, vivo experiments significantly suppressed cartilage tissue damage, ferroptosis, IHC scores CYP1B1 rats. conclusion, our results suggested KLF9, epigenetic silenced DNMT1, promoted extracellular signal‐regulated kinase (ERK)‐mediated through transcriptionally regulating CYP1B1. Thus, expected new target for treatment OA.

Autophagy is responsible for maintaining cellular balance and ensuring survival. plays a crucial role in the development of diseases, particularly human cancers, with actions that can either promote survival or induce cell death. However, brain tumors contribute to high levels both mortality morbidity globally, resistance treatments being acquired due genetic mutations dysregulation molecular mechanisms, among other factors. Hence, having knowledge processes advancement enlightening, current review specifically examines autophagy. The discussion would focus on pathways control autophagy tumors, its dual as tumor suppressor supporter different types like glioblastoma, glioma, ependymoma, demonstrating potential treatment. mechanisms influence metastasis drug there complex interplay between responses stress hypoxia starvation. inhibit growth by promoting apoptosis. focusing could offer fresh perspectives creating successful treatments.

Abstract Glioma is the most common malignant tumor in central nervous system, with significant health burdens to patients. Due intrinsic characteristics of glioma and lack breakthroughs treatment modalities, prognosis for patients remains poor. This results a heavy psychological financial load worldwide. In recent years, cannabidiol (CBD) has garnered widespread attention research due its anti-tumoral, anti-inflammatory, neuroprotective properties. review comprehensively summarizes preclinical clinical on use CBD therapy, as well current status nanomedicine formulations CBD, discusses potential challenges therapy future.
