Dosing zero valent iron (ZVI) or sodium hydroxide (NaOH) is the common method of addressing acidification in anaerobic digestion (AD) systems; however, few studies have discussed and compared their effects on microbial metabolism. In present study, syntrophy metabolic pathways under ZVI NaOH regulation are comparatively analyzed through network analysis metagenomic/metaproteomic analyses. CH4 yield reactor was 414 mL/gVS, an increase 23% when with that dosing (336 mL/gVS). The methanogenesis recovery period (37 days) shorter than (48 days). Co-occurrence networks indicated promoted Methanoculleus Methanosarcina to establish a complex syntrophic association SAO bacteria (Syntrophaceticus Aminobacterium) acetogens (Syntrophomonas), strengthening SAO-hydrogenotrophic (HM) acetoclastic (AM) simultaneously. Metagenomic showed relative abundance mcrA fwdB higher 27% reactor. Furthermore, metaproteomics analysis, much more enzymes related glucose degradation, bioconversion butyric acid pyruvate, conversion formate acetate CO2, production from CO2 were significantly upregulated (fold change control [FC] > 1.5, p < 0.05). results study enhance our understanding methanogenic mechanisms ZVI, providing theoretical basis for its practical application AD systems experiencing VFA suppression.
