Mitochondria are important organelles that provide energy for cellular physiological activities. Changes in their structures may indicate the occurrence of diseases, and super-resolution imaging mitochondria is great significance. However, developing fluorescent probes mitochondrial visualization still remains challenging due to insufficient fluorescence brightness poor stability. Herein, we rationally synthesized an ultrabright xanthene probe Me-hNR mitochondria-specific using structured illumination microscopy (SIM). The rigid structure provided its ultrahigh quantum yield up 0.92 16,000. Occupying para-position O atom skeleton by utilizing smallest methyl group ensured excellent study photophysical process indicated mainly emitted via radiative decay, nonradiative decay inter-system crossing were rare slow rate large gap (ΔEst = 0.55 eV). Owing these merits, can specifically light at ultralow concentrations down 5 nM. unprecedented spatial resolution with fwhm 174 nm was also achieved. Therefore, this has potential visualizing structural changes revealing pathogenesis related diseases SIM.
