Dry Eye Disease (DED) significantly impacts global populations, causing discomfort and vision problems. This review explores the effects of lutein supplementation on DED symptoms signs. A systematic was conducted following PRISMA guidelines, examining clinical trials from databases including PubMed, Web Science, EMBASE, Cochrane Library. Six randomized controlled (RCTs) involving 584 subjects were included. Meta-analysis not due to heterogeneity in study designs, dosages, outcome measures. Lutein dosages ranged 3 mg/day 20 mg/day, with treatment durations 4 12 weeks. Improvements observed subjective symptoms, significant reductions Ocular Surface Index (OSDI) scores some studies. Objective measures also showed positive results: tear break-up time (TBUT) increased trials. However, other studies reported no differences between control groups, reflecting outcomes. Schirmer's test corneal-conjunctival staining results varied, showing improvements others not. may benefit patients by improving film stability. heterogeneity, larger, well-designed RCTs are needed establish standardized dosing confirm these findings.

Drug-delivery systems (DDSs) are designed to deliver drugs their specific targets minimize toxic effects and improve susceptibility clearance during targeted transport. Peptides have high affinity, low immunogenicity, simple amino acid composition, adjustable molecular size; therefore, most peptides can be coupled via linkers form peptide-drug conjugates (PDCs) act as active pro-drugs. PDCs widely thought promising DDSs, given ability drug bio-compatibility physiological stability. Peptide-based DDSs often used therapeutic substances such anti-cancer nucleic acid-based drugs, which not only slow the degradation rate of in vivo but also ensure concentration at site prolong half-life vivo.This article provides an profile advancements future development functional peptide-based delivery both domestically internationally recent years, expectation achieving incorporating taking full advantage synergistic effects.

Abstract Oral drug delivery is a promising approach for the treatment of various gastrointestinal (GI) diseases but poses significant challenges to target non‐GI diseases. The intestinal barrier forms anatomical challenge reach diseased site, along with physiological such as stability in GI tract. These lead development targeted nanoparticles and strategies cross protect them from harsh conditions tract, improving absorption into circulatory system, bioavailability, ensuring regulated release. Targeting ligands chitosan, Butyrate (BU), yeast capsule (YC) shows effective permeability across epithelium. After crossing epithelium, these can effectively treat atherosclerosis, cancers, neurodegenerative diseases, fibrosis, post‐traumatic osteoarthritis. However, including low bioavailability still persist, which reduce efficacy therapeutics should be considered designing potential therapies near future.

Strong precorneal clearance mechanisms including reflex blink, constant tear drainage, and rapid mucus turnover constitute great challenges for eye drops effective drug delivery to the ocular epithelium. In this study, cyclosporine A (CsA) treatment of dry disease (DED) was selected as model drug. Two strategies, PEGylation penetration cationization potent cellular uptake, were combined construct a novel CsA nanosuspension (NS@lipid-PEG/CKC) by coating nanoscale particles with mixture lipids, DSPE-PEG2000, cationic surfactant, cetalkonium chloride (CKC). NS@lipid-PEG/CKC mean size ∼173 nm positive zeta potential ∼+40 mV showed promoted penetration, good cytocompatibility, more prolonged retention without obvious irritation. More importantly, recovered production goblet cell density efficiently than commercial nanoemulsion on rat model. All results indicated that combination might provide promising strategy coordinate uptake enhanced epithelium nanomedicine-based drops.
