Cancer is a complex genetic disorder characterized by abnormalities in both coding and regulatory non-coding RNAs. microRNAs (miRNAs) are key RNAs that modulate cancer development, functioning as tumor suppressors oncogenes. miRNAs play critical roles progression, influencing processes such initiation, promotion, metastasis. They exert their effects targeting suppressor genes, thereby facilitating while also inhibiting oncogenes to prevent further disease advancement. The miR-10 family, particularly miR-10a-5p miR-10b-5p (miR-10a/b-5p), notably involved progression. Intriguingly, functions can differ across different cancers, sometimes promoting at other times suppressing growth depending on the type target genes. This review explores dual of miR-10a/b-5p tumor-suppressive (TSmiRs) or oncogenic (oncomiRs) various cancers examining molecular cellular mechanisms impact microenvironment. Furthermore, we discuss potential therapeutic targets, emphasizing miRNA-based strategies for treatment. insights discussed this aim advance our understanding miR-10a/b-5p’s biology application developing innovative therapies.

Sepsis is a life-threatening syndrome characterized by organ dysfunction caused dysregulated host response to infection. Sepsis-associated acute liver injury (SA-ALI) frequent and serious complication of sepsis that considerably impacts both short-term long-term survival outcomes. In intensive care units (ICUs), the mortality rate patients with SA-ALI remains high, mostly due absence effective early diagnostic markers suitable therapeutic strategies. Recent studies have demonstrated importance non-coding RNAs (ncRNAs) in development progression SA-ALI. This review focuses on critical roles ncRNAs, including microRNAs (miRNAs), long (lncRNAs), circular (circRNAs), regulating "cytokine storms", oxidative stress, mitochondrial dysfunction, programmed cell death SA-ALI, summarizes current state limitations existing lncRNAs circRNAs By integrating advancements high-throughput sequencing technologies, this provides novel insights into dual potential ncRNAs as biomarkers targets, offers new ideas for diagnosis treatment research highlights challenges clinical translation.

Long non-coding RNAs (lncRNAs) have been recognized for their crucial roles in the replication processes of various viruses. However, specific functions and regulatory mechanisms many lncRNAs influenza A virus (IAV) pathogenesis remain poorly understood. In this study, we identified lncRNA THRIL observed a significant reduction its expression following IAV infection A549 cells. The treatment cells with viral mimic poly (I:C), or type I III interferons, resulted substantial decrease expression. Furthermore, overexpression significantly enhanced replication, while silencing markedly reduced replication. Additionally, led to notable reductions levels interferons cell lines overexpressing compared control groups; conversely, knockdown exhibited higher interferon than groups. Moreover, was found inhibit several critical interferon-stimulated genes (ISGs), which are essential an effective antiviral response. Notably, our findings demonstrated that impaired activation IRF3, key transcription factor signaling pathway, thereby suppressing host innate immunity. These results highlight THRIL’s potential as therapeutic target strategies.

Abstract Motivation Despite significant advances in spatial transcriptomics, the analysis of formalin-fixed paraffin-embedded (FFPE) tissues, which constitute most clinically available samples, remains challenging. Additionally, capturing both coding and noncoding RNAs a context poses challenges. We recently introduced Patho-DBiT, technology designed to address these unmet needs. However, marked differences between Patho-DBiT existing transcriptomics protocols necessitate specialized computational tools for comprehensive whole-transcriptome FFPE samples. Results Here, we present ASTRO, an automated pipeline developed process data. In addition supporting standard datasets, ASTRO is optimized analyses enabling detection various RNA species, including non-coding such as miRNAs. To compensate reduced quality incorporates filtering step optimizes barcode calling, increasing mapping rate. These optimizations allow spatially quantify species entire transcriptome achieve robust performance Availability Codes are at GitHub ( https://github.com/gersteinlab/ASTRO ).

Non-coding ncRNAs (ncRNAs) are transcriptional production which do not coding proteins, with a spectrum of diversified type, and function (Chen Kim, 2024). Although the have many types including long noncoding RNAs (lncRNAs), microRNAs (miRNAs), small nucleolar (snoRNAs), transfer RNA (tRNA), ribosomal (rRNA), circular (circRNA) etc., most them lncRNA miRNA. In 1984, first regulatory ncRNA, microRNA F (micF) was discovered from E. coli. MicF inhibits translation OmpF by binding to its ribosome-binding site, blocking ribosome entry (Mizuno et al., 1984;Corcoran 2012).Over past 40 years, numerous non-coding (ncRNAs), such as Let-7, enod40, H19, NEAT1, been identified key regulators in animals or plants (Bartolomei 1991;Crespi 1994;Pasquinelli 2000;Hutchinson 2007).
