Epstein-Barr virus (EBV) causes infectious mononucleosis, triggers multiple sclerosis, and is associated with 200,000 cancers/year. EBV colonizes the human B cell compartment periodically reactivates, inducing expression of 80 viral proteins. However, much remains unknown about how remodels host cells dismantles key antiviral responses. We therefore created a map EBV-host EBV-EBV interactions in undergoing replication, uncovering conserved herpesvirus versus EBV-specific targets. The EBV-encoded G-protein-coupled receptor BILF1 MAVS UFM1 E3 ligase UFL1. Although UFMylation 14-3-3 proteins drives RIG-I/MAVS signaling, BILF1-directed instead triggered packaging into mitochondrial-derived vesicles lysosomal proteolysis. In absence BILF1, replication activated NLRP3 inflammasome, which impaired pyroptosis. Epstein-Barr virus (EBV) is a ubiquitous human lymphotropic herpesvirus that causally associated with several malignancies. In addition to latent factors, lytic replication contributes cancer development. this study, we examined whether the gene BNRF1, which conserved among gamma-herpesviruses, has an important role in lymphomagenesis. We found lymphoblastoid cell lines (LCLs) established by BNRF1-knockout EBV exhibited remarkably lower pathogenicity mice xenograft model than LCLs produced wild-type (LCLs-WT). RNA-seq analyses revealed BNRF1 elicited expression of interferon-inducible protein 27 (IFI27), promotes proliferation. IFI27 knockdown LCLs-WT resulted excessive production reactive oxygen species, leading death and significantly decreased their vivo . also confirmed was upregulated during primary infection B-cells. Abstract Nuclear pore complexes (NPCs) have increasingly recognized interactions with the genome, as exemplified in yeast, where they bind transcribed or damaged chromatin. By combining genome-wide approaches live imaging of model loci, we uncover a correlation between NPC association and accumulation R-loops, which are genotoxic structures formed through hybridization nascent RNAs their DNA templates. Manipulating hybrid formation demonstrates that R-loop per se, rather than transcription R-loop-dependent damages, is primary trigger for relocation to NPCs. Mechanistically, repositioning involves recognition by ssDNA-binding protein RPA, SUMO-dependent NPC-associated factors. Preventing leads lethality hybrid-accumulating conditions, while tethering hybrid-prone locus attenuates genetic instability. Remarkably, this pathway molecular factors similar those required stalled replication forks NPCs, supporting existence convergent mechanisms sensing transcriptional stresses.
