Abstract In recent years, there has been a growing interest in understanding the function of claustrum. The claustrum is thin, long, subcortical structure with dense connections to cortex. Despite these extensive connections, manner which influences broader cortical activity remains unclear. We used mesoscale calcium fluorescence imaging examine resting state (1-5Hz) and sensory-evoked responses lightly anesthetized mice, before after bi-directional chemogenetic modulation claustrum-prefrontal neurons. The claustrum (CLA) is a conspicuous subcortical structure interconnected with cortical and regions. Its regional anatomy cell-type-specific connections in the mouse remain not fully determined. Using multimodal reference datasets, we confirmed delineation of CLA as single group neurons embedded agranular insular cortex. Abstract The claustrum has been linked to attention and sleep. We hypothesized that this reflects a shared function, determining responsiveness stimuli, which spans the axis of engagement. To test hypothesis, we recorded population dynamics from male mice during both sleep an attentional task (‘ENGAGE’). Heightened activity in neurons projecting anterior cingulate cortex (ACCp) corresponded reduced sensory Similarly, ENGAGE task, heightened ACCp correlated with disengagement behavioral lapses, while low hyper-engagement impulsive errors. Abstract The anatomical organization of the rodent claustrum remains obscure due to lack clear borders that distinguish it from neighboring forebrain structures. Defining what constitutes is imperative for elucidating its functions. Methods based on gene/protein expression or transgenic mice have been used spatially outline but often report incomplete labeling and/or specificity during certain neurodevelopmental timepoints. To reliably identify projection cells in mice, we propose a simple immunolabelling method juxtaposes pattern claustrum-enriched and cortical-enriched markers. We determined immunoreactive markers Nurr1 Nr2f2 are devoid cortical marker Tle4, which allowed us differentiate adjoining cells. Using retrograde tracing, verified nearly all neurons Tle4 expressed Nurr1/Nr2f2 different degrees. At neonatal stages between 7 21 days, were identified by their Nurr1-postive/Tle4-negative profile, time-period when other techniques localize adult ineffective. Finally, exposure environmental novelty enhanced neuronal activation c-Fos region. Notably, was mainly restricted Nurr1-positive absent Tle4-positive cells, thus corroborating previous work reporting novelty-induced activation. Taken together, this will aid studying postnatal development may improve histological functional studies where approaches not amenable.

Non-human studies suggest that the claustrum, small subcortical nucleus, coordinates slow waves. Abstract The claustrum (CLA) is a brain nucleus located between the insula and lateral striatum, implicated in wide range of behaviors. Underpinning different behavioral phenotypes connectivity various cortical regions, including anterior cingulate cortex (ACC). CLA projection neurons are glutamatergic neurons, however, impact on its targets has been shown some studies to be inhibitory. Such inhibition likely arise from claustral activation interneurons, intricate synaptic cell types not known. Here, we combine vivo ex electrophysiology optogenetics reveal functional organization CLA-ACC pathway according identity pre- postsynaptic populations. Optogenetic stimulation awake mice resulted multiphasic excitatory inhibitory responses ACC cells, which depended layer, type, stimulated population.
