Abstract The Drosophila melanogaster (fruit fly) misexpression suppressor of Ras 4 (MESR4) gene encodes a potential transcription factor and plays critical roles in various biological processes, including embryonic development, lipid metabolism, eye–antennal germline stem cell differentiation. However, whether it is involved modulating intestinal homeostasis remains elusive. In this study, we provide compelling evidence demonstrating that MESR4 bona fide regulator preventing age‐onset leakage dysbiosis adult flies. Mechanistically, predominantly located at the nucleus cells controls expression bag‐of‐marbles ( bam ), thereby restricting excessive activation immune deficiency signaling during aging. silencing Relish Rel which key pathway, reverses beneficial effects mediating barrier function fly lifespan. Collectively, our studies uncover an undescribed maintenance overall organismal fitness.

Cullin 2 (Cul2), a core component of the Cullin-RING E3 ubiquitin ligase complex, is integral to regulating distinct biological processes. However, its role in innate immune defenses remains poorly understood. In this study, we investigated functional significance Cul2 deficiency (IMD) signaling-mediated antimicrobial reactions Drosophila melanogaster (fruit fly). We demonstrated that loss-of-function led marked reduction peptide induction following bacterial infection, which was associated with increased fly mortality and load. The proteomic analysis further revealed reduced expression Effete (Eff), key E2 ubiquitin-conjugating enzyme during IMD signaling. Intriguingly, ectopic eff effectively rescued defects caused by loss Cul2. Taken together, results our study underscore critical ensuring robust signaling activation, highlighting importance defense against microbial infection Drosophila.
