Pacinian corpuscles detect transient touch and vibration in vertebrates. Corpuscles are composed of a mechanoreceptor afferent surrounded by lamellar Schwann cells (LSCs), enclosed multilayered outer core. The spatial arrangement these components their contribution to sensory tuning unclear. We report the three-dimensional architecture corpuscle reveal role its cellular detection. In prevailing model, core acts as mechanical filter that limits static low-frequency stimuli from reaching terminal—the presumed sole site show is dispensable for vibration; instead, properties arise inner By acting additional sensors, LSCs potentiate mechanosensitivity terminal, which detects via fast inactivating ion channels. Thus, functional enabled an interplay between mechanosensitive terminal

Summary Pacinian corpuscles are rapidly adapting mechanoreceptor end-organs that detect transient touch and high-frequency vibration. In the prevailing model, these properties determined by outer core, which acts as a mechanical filter limiting static low-frequency stimuli from reaching afferent terminal—the sole site of detection in corpuscles. Here, we determine detailed 3D architecture corpuscular components reveal their contribution to detection. We show core is dispensable for rapid adaptation frequency tuning. Instead, arise inner composed gap junction-coupled lamellar Schwann cells (LSCs) surrounding terminal. By acting additional sensing structures, LSCs potentiate mechanosensitivity terminal, detects via fast-inactivating ion channels. propose model corpuscle function mediated an interplay between mechanosensitive terminal core. Mechanosensitive Piezo channels are ion activated by mechanical stimuli, playing a crucial role in mechanotransduction processes and hypersensitivity. When these subjected to loading, membrane currents rise instantaneously, depolarizing activating voltage-gated calcium channels. This results an increase intracellular Ca
