The
reprogramming
of
metabolic
pathways
and
processes
in
immune
cells
has
emerged
as
an
important
aspect

response.
Metabolic
intermediates
accumulate

In
macrophages,
there
been
major
focus
on
3
metabolites
linked
to
Krebs
cycle,
itaconate,
succinate,
fumarate,
which
have
shown
regulate
multiple
processes.
Here,
we
discuss
recent
progress
these
with
regard
their
effect
macrophages
host
defense
inflammatory
diseases.
We
also
consider
therapeutic
opportunities
presented
from
mimicry
or
by
targeting
enzymes
that
make
metabolize
them
order
leverage
body's
own
anti-inflammatory

We
have
developed
two
monoclonal
antibodies,
CPTC-2MeSC-1
and
CPTC-2MeSC-2,
against
itaconate
its
conjugates
with
sulfhydryl-containing
biomolecules
such
as
cysteines.
Itaconate
is
a
dicarboxylic
acid
metabolite
that
has
recently
gained
much
interest
for
anti-inflammatory
properties
in
many
biological
models.

ITA-Cys
designed
to
mimic
vivo

adducts
of
itaconate.
Two
antibodies
ITA-Cys,
CPTC-2MeSC,
were
shown
high
immunoreactivity
unconjugated
itaconate,

conjugates,
dimethyl
found
CPTC-2MeSC-2
are
specific

bind
other
structurally
similar
cysteine
adducts,
including
those
obtained

metabolites
