Abstract Metal‐organic frameworks (MOFs) are a new variety of solid crystalline porous functional materials. As an extension inorganic materials, it has made important progress in preparation and application. MOFs widely used various fields such as gas adsorption storage, drug delivery, sensing, biological imaging due to their high specific surface area, porosity, adjustable pore size, abundant active sites, modification by introducing groups. In this paper, the types classified, synthesis methods mechanisms materials summarized. Finally, application prospects challenges metal‐organic framework biomedical field discussed, hoping promote multidisciplinary fields.

Nanotechnology, particularly quantum dots (QDs), has ushered in a transformative era the pharmaceutical and medical industries, offering notable opportunities for nanoscale advancements. These particles, known their exceptional optical properties confinement, have emerged as indispensable tools cancer drug delivery bioimaging. This review delves into various conjugation techniques with QDs, including covalent linking, non-covalent conjugation, click chemistry, disulfide linkage, pH-sensitive linkage. Each method provides distinct advantages, such enhanced stability, reversibility, specificity, controlled release. Moreover, QDs demonstrated significant promise oncology by efficiently delivering drugs to cancerous tissues while minimising systemic toxicity. Investigations applications different cancers, blood, brain, cervical, breast etc., reveal efficacy targeted delivery, real-time imaging, improved therapeutic outcomes. However, challenges potential toxicity, pharmacokinetics, targeting specificity must be addressed fully harness benefits of therapy. Future research should focus on developing biocompatible optimising techniques, elucidating safety profiles long-term effects biological systems. Overall, represent promising frontier treatment, multifaceted capabilities that hold outcomes reduced side across cancers.

The development of nanoparticle (NP)-based drug carriers has presented an exciting opportunity to address challenges in oncology. Among the 100,000 available possibilities, zirconium-based metal–organic frameworks (MOFs) have emerged as promising candidates biomedical applications. Zr-MOFs can be easily synthesized small-size NPs compatible with intravenous injection, whereas ease decorating their external surfaces functional groups allows for targeted treatment. Despite these benefits, suffer degradation and aggregation real, vivo conditions, loaded drugs will burst effect─i.e., fast release less than 48 h. To tackle issues, we developed a simple but effective bilayer coating strategy generic, two-step process. Abstract Despite improvements in cancer survival rates, metastatic and surgery‐resistant cancers, such as pancreatic cancer, remain challenging, with poor prognoses limited treatment options. Enhancing drug bioavailability tumors, while minimizing off‐target effects, is crucial. Metal–organic frameworks (MOFs) have emerged promising delivery vehicles owing to their high loading capacity, biocompatibility, functional tunability. However, the vast chemical diversity of MOFs complicates rational design biocompatible materials. This study employed machine learning molecular simulations identify suitable for encapsulating gemcitabine, paclitaxel, SN‐38, identified PCN‐222 an optimal candidate. Following loading, MOF formulations are improved colloidal stability biocompatibility. In vitro studies on cell lines shown cellular internalization, delayed release. Long‐term tests demonstrated a consistent performance over 12 months. vivo tumor‐bearing mice revealed that paclitaxel‐loaded PCN‐222, particularly hydrogel local administration, significantly reduced spread tumor growth compared free drug. These findings underscore potential effective system hard‐to‐treat cancers.
