Two-dimensional nanomaterials (2DNMs) exhibit significant potential for the development of functional and specifically targeted biosensors, owing to their unique planar nanosheet structures distinct physical chemical properties. Biomodification biomimetic synthesis offer green mild approaches fabrication multifunctional nanohybrids with enhanced catalytic, fluorescent, electronic, optical properties, thereby expanding utility in constructing high-performance biosensors. In this review, we present recent advances 2DNM-based via both biomodification strategies biosensor applications. We discuss covalent non-covalent methods involving various biomolecules, including peptides, proteins, DNA/RNA, enzymes, biopolymers, bioactive polysaccharides. The engineering biomolecule–nanomaterial interfaces creation biomodified is also explored. Furthermore, summarize bio–nanohybrids through pathways such as bio-templating, biomolecule-directed self-assembly, biomineralization, integration. applications these diverse biosensing platforms—including colorimetric, surface plasmon resonance, electrochemical, fluorescence, photoelectrochemical, integrated multimodal biosensors—are introduced discussed.
