Shift work increases the risk for human errors, such that drowsiness due to shift has contributed major industrial disasters, including Space Shuttle Challenger, Chernobyl and Alaska Oil Spill with extraordinary socio-economical costs. Overnight operations pose a challenge because our circadian biology inhibits cognitive performance at night. Yet how system modulates cognition over multiple days under realistic conditions remains be established. Importantly, task-specific brain regions show different 24-h dynamics, we hypothesize misalignment impacts task-dependently. Using biologically-driven paradigm mimicking night work, randomized, cross-over design, between pacemaker behavioral/environmental cycles vulnerability on sustained attention, throughput, information processing visual-motor days, compared alignment (day shifts). Circadian effects are task-dependent: while they acutely impair attention recovery after 3-days, progressively hinder daily learning. Individuals felt sleepier during misalignment, but did not rate their as worse. Furthermore, depended prior sleep history. Abstract Circadian clocks are endogenous oscillators that control 24-h physiological and behavioral processes. The central circadian clock exerts over myriad aspects of mammalian physiology, including the regulation sleep, metabolism, immune system. Here, we review advances in understanding genetic sleep through system, as well impact dysregulated gene expression on metabolic function.
