Abstract Background Endoscopic submucosal dissection (ESD) is the current standard treatment for early-stage esophageal neoplasms. However, postoperative stricture after extensive mucosal remains a severe challenge with limited effective treatments available. In this study, we introduced chitosan/gelatin (ChGel) sponge encapsulating adipose mesenchymal stem cells (ADMSCs)-derived exosomes (ChGel MSC−Exo ) prevention of stenosis ESD in porcine model. Results Pigs were randomly assigned into (1) ChGel group, (2) PBS and (3) controls. Exosome applied immediately on day as well 7. components crucial wound healing investigated by liquid chromatography-tandem mass spectrometry (LC–MS/MS) small RNA sequencing. significantly reduced contraction 21, less fiber accumulation inflammatory infiltration, enhanced angiogenesis when compared control groups. The anti-fibrotic effects following MSC-Exo further found to be associated anti-inflammatory M2 polarization resident macrophages, especially within M2b subset characterized TGFβ1 secretion, which sufficiently inhibited inflammation prevented activation myofibroblast collagen production at early stage ESD. Moreover, abundant expression exosomal MFGE8 was identified involved transition M2b-macrophage through MFGE8/STAT3/Arg1 axis. Conclusions Our study demonstrates that promotes subset, consequently reducing deposition. These findings suggest promising potential therapy preventing development near-circumferential Graphical

Abstract Macrophages exist in various tissues, several body cavities, and around mucosal surfaces are a vital part of the innate immune system for host defense against many pathogens cancers. possess binary M1/M2 macrophage polarization settings, which perform central role an array tasks via intrinsic signal cascades and, therefore, must be precisely regulated. Many crucial questions about signaling modulation yet to uncovered. In addition, clinical importance tumor-associated macrophages is becoming more widely recognized as significant progress has been made understanding their biology. Moreover, they integral tumor microenvironment, playing regulation wide variety processes including angiogenesis, extracellular matrix transformation, cancer cell proliferation, metastasis, immunosuppression, resistance chemotherapeutic checkpoint blockade immunotherapies. Herein, we discuss signaling, mechanical stresses modulation, metabolic pathways, mitochondrial transcriptional, epigenetic regulation. Furthermore, have broadly extended traps essential roles autophagy aging regulating functions. discussed recent advances macrophages-mediated autoimmune diseases tumorigenesis. Lastly, targeted therapy portray prospective targets therapeutic strategies health diseases.

Abstract Psoriasis is a common, chronic, and inflammatory skin disease with high burden on individuals, health systems, society worldwide. With the immunological pathologies pathogenesis of psoriasis becoming gradually revealed, therapeutic approaches for this have gained revolutionary progress. Nevertheless, mechanisms less common forms remain elusive. Furthermore, severe adverse effects recurrence upon treatment cessation should be noted addressed during treatment, which, however, has been rarely explored integration preliminary findings. Therefore, it crucial to comprehensive understanding behind pathogenesis, which might offer new insights research lead more substantive progress in expand clinical options treatment. In review, we looked briefly introduce epidemiology, subtypes, pathophysiology, comorbidities systematically discuss signaling pathways involving extracellular cytokines intracellular transmission, as well cross-talk between them. discussion, also paid attention potential metabolic epigenetic molecular mechanistic cascades related its comorbidities. This review outlined current psoriasis, especially targeted therapies novel strategies, mechanism recurrence.

Exosomes are small extracellular vesicles that carry lipids, proteins, and microRNAs (miRNAs). They released by all cell types can be found not only in circulation but many biological fluids. essential for interorgan communication because they transfer their contents from donor to recipient cells, modulating cellular functions. The miRNA content of exosomes is responsible most effects, changes exosomal levels contribute the progression or regression metabolic diseases. As miRNAs selectively sorted packaged into exosomes, useful as biomarkers diagnosing field metabolism expanding rapidly, researchers consistently making new discoveries this area. a result, have great potential next-generation drug delivery platform

Abstract Sepsis, a severe systemic inflammatory response to infection, remains leading cause of morbidity and mortality worldwide. Exosomes, as mediators intercellular communication, play pivotal role in the pathogenesis sepsis through modulating immune responses, metabolic reprogramming, coagulopathy, organ dysfunction. This review highlights emerging significance exosomes these processes. Initially, it provides an in-depth insight into exosome biogenesis characterization, laying groundwork for understanding their diverse intricate functions. Subsequently, explores regulatory roles various cells such neutrophils, macrophages, dendritic cells, T B cells. analysis elucidates how are thus contributing complexity pathophysiology. Additionally, this delves regulation metabolism subsequent dysfunction sepsis. It also establishes connection between coagulation cascade, which affects endothelial integrity promotes thrombogenesis Moreover, discusses dual progression resolution sepsis, exploring complex involvement inflammation healing Furthermore, underscores potential biomarkers therapeutic targets. Understanding mechanisms presents new opportunities novel interventions mitigate outcomes emphasizing promise research critical care settings.

The global epidemic of metabolic diseases has led to the emergence dysfunction-associated steatotic liver disease (MASLD) and steatohepatitis (MASH), which pose a significant threat human health. Despite recent advances in research on pathogenesis treatment MASLD/MASH, there is still lack more effective targeted therapies. Extracellular vesicles (EVs) discovered wide range tissues body fluids encapsulate different activated biomolecules mediate intercellular communication. Recent studies have shown that EVs derived from adipose tissue (AT) play vital roles MASLD/MASH therapeutics, depending their sources intervention types. Besides, adipose-derived stem cell (ADSC)-derived appear be mitigating MASLD/MASH. This review presents an overview definition, extraction strategies, characterisation EVs, with particular focus biogenesis release exosomes. It also reviews effects potential molecular mechanisms liver- AT-derived emphasises contribution clinical therapeutic ADSC-derived EVs. Furthermore, future perspective EV therapy setting discussed.
