The gut microbiota can produce a variety of microbial-derived metabolites to influence tumor development. Tryptophan, an essential amino acid in the human body, be converted by microorganisms via indole pathway such as Indole-3-Lactic Acid (ILA), Indole-3-Propionic (IPA), Indole Acetic (IAA) and Indole-3-Aldehyde (IAld). Recent studies have shown that play key roles progression, they used adjuvant regimens for immunotherapy or chemotherapy. Here, we summarize recent findings on common microbial provide review mechanisms different microenvironment. We further discuss limitations current metabolite research future possibilities. It is expected will new strategies clinical therapy.

Abstract Ischemic stroke is the most common cerebrovascular disease and leading cause of permanent disability worldwide. Recent studies have shown that development prognosis are closely related to abnormal tryptophan metabolism. Here, significant downregulation 3‐hydroxy‐kynurenamine (3‐HKA) in patients animal models identified. Supplementation with 3‐HKA improved long‐term neurological recovery, reduced infarct volume, increased ipsilateral cerebral blood flow after distal middle artery occlusion (MCAO). promoted angiogenesis, functional vessel formation, blood‐brain barrier (BBB) repair. Moreover, inhibited A1‐like (neurotoxic) astrocyte activation but A2‐like (neuroprotective) polarization. Proteomic analysis revealed AIM2 inflammasome stroke, co‐labeling indicated expression typically astrocytes at 7 14 days stroke. Consistently, co‐cultures primary mouse brain microvascular endothelial cells astrocytes, angiogenesis oxygen‐glucose deprivation (OGD). overexpression abrogated 3‐HKA‐driven vascular remodeling vitro vivo, suggesting may regulate astrocyte‐mediated by impeding activation. In conclusion, promote post‐stroke regulating A1/A2 activation, thereby improving supplementation be an efficient therapy for

The bidirectional relationship between the gut microbiota and nervous system is known as microbiota-gut-brain axis (MGBA). MGBA controls complex interactions brain, enteric system, gut-associated immune neuroendocrine systems, regulating key physiological functions such response, sleep, emotions mood, food intake, intestinal functions. Psychobiotics are considered tools with potential to modulate through preventive, adjunctive, or curative approaches, but their specific mechanisms of action on many aspects health yet be characterized. This narrative review perspectives article highlights paradigms needing attention scope probiotics applications in human increases, a growing body evidence supporting systemic beneficial effects. However, there limitations overcome before establishing extent which we can incorporate management neuropsychiatric disorders. Although this uses term general manner, it remains important study at strain level most cases.

Abstract The progression of colorectal cancer is closely associated with diet. Fasting-mimicking diet (FMD) a promising type dietary intervention that have beneficial effects in the prevention and treatment various cancers. We investigated therapeutic effect 4-day FMD against mice through immune cell analysis, microbiota composition analysis anti-PD-1 treatment. These cycles effectively suppressed growth, reduced proliferation angiogenesis, increased tumor-infiltration lymphocytes especially CD8 + T cells. stimulated protective gut microbiota, Lactobacillus . Supplementation johnsonii induced similar results as intervention, which also tumor growth CD45 Additionally, synthesizing therapy inhibited CRC progression. findings suggest Lactobacillus. necessary for anticancer process CRC. its on both system, mouse model.

Abstract High myopia (HM) is a leading cause of blindness worldwide with currently no effective interventions available. A major hurdle lies in its often isolated perception as purely ocular morbidity, disregarding potential systemic implications. Recent evidence suggests the existence gut-eye axis; however, role gut microbiota pathogenesis HM remains largely unexplored. Herein, we provide crosstalk among HM’s dysbiosis, microbial metabolites, and scleral remodeling. Utilizing 16S rRNA gene sequencing, observed an altered profile patients significant reduction probiotic abundance compared healthy controls. Subsequent targeted metabolic profiling revealed notable decrease plasma levels microbiota-derived metabolite indole-3-acetic acid (3-IAA) patients, which closely associated reduced probiotics, both negatively correlated severity. Genetic analyses determined that are causally risk. Importantly, when mice subjected to modeling receive fecal transplantation from donors, there increase 3-IAA simultaneous retardation progression along better maintenance collagen type I alpha 1 (COL1A1) expression sclera. Furthermore, gavage achieves similar effects. Mechanistic investigations confirm transcriptional activation COL1A1 by via promoting enrichment SP1 promoter. Together, our findings novel insights into microbiota-eye axis propose new strategies for intervention remodeling indole supplementation.

Chronic stress is closely associated with gastrointestinal disorders. However, the impact of stress-related neurotransmitters such as serotonin (5-hydroxytryptamine, 5-HT) on intestines under chronic conditions remains poorly understood. This study aims to elucidate mechanisms by which 5-HT affects mitochondrial biogenesis and intestinal barrier integrity during stress. Employing a restraint (CRS) mouse model, we observed elevated levels, altered colonic mucosal structure, disrupted tight junctions. The increase in was up-regulated synthesis enzymes downregulated reuptake transporters, indicating an imbalance homeostasis caused Furthermore, exacerbated oxidative impaired junction protein expression, highlighting its role promoting dysfunction. Background Autism spectrum disorder (ASD) is an early-onset neurodevelopmental disorder, usually accompanied by gut microbiota dysregulation. Gut homeostasis considered effective for ASD. Reportedly, Dachaihu decoction (DCHD) can efficiently regulate and inflammation. However, the mechanisms underlying effects of DCHD in treatment ASD remain unclear. Objective This study investigated potential treating Methods In animal experiment, propionic acid was administered to construct rat model. The rats were treated with DCHD, efficacy assessed using behavioral detections, such as open field test, elevated plus maze novel object recognition test. Additionally, levels IL-6, TNF- α , IL-10, T-SOD, MDA, GSH CAT determined kits, histological staining used evaluate brain morphology. Moreover, tight junction proteins (ZO-1 occludin) expression evaluated RT-qPCR, whereas Iba1 level immunofluorescence staining. 16S rRNA sequencing metabolomic analysis feces revealed targets against a small human trail, clinical scales ADOS-2 Behavior Checklist (ABC) autism severity. Gastrointestinal problems function based on food intolerance event-related potential, respectively. Results significantly improved autism-like behaviors increased antioxidant enzyme activity, decreased inflammation enhanced intestinal barrier experiment. Furthermore, altered profile, probiotics Adlercreutzia Parvibacter Turicibacter Christensenellaceae. Further, beneficial metabolite indole-3-acetate cognitive impairment-related metabolites asymmetric dimethylarginine homogentisic acid. Meanwhile, trial that alleviated core symptoms ASD, ABC scale scores. also specific egg white/yolk milk IgG antibodies shortened MMN P3b latencies. Conclusion demonstrated may alleviate via inhibiting oxidative stress, reducing inflammation, modulating rats.
