Abstract: Studies examining post-traumatic stress disorder (PTSD) have either emphasized a relationship between PTSD and systemically pro-inflammatory state or identified link chronic disease. The aim of this study was to evaluate the evidence for individuals with systemic low-grade inflammation that has been proposed underlie disease development in population. authors conducted systematic review literature (January 2006 April 2017) accordance PRISMA statement following four databases: PubMed, MEDLINE, PsycINFO, SPORTDiscus Full Text. search strategy limited articles published peer-reviewed journals human studies. Posttraumatic stress disorder (PTSD) is a chronic impairment that occurs after exposure to traumatic events. This can result in disturbance individual and family functioning, causing significant medical, financial, social problems. study selective review of literature aiming provide general outlook the current understanding PTSD. There are several diagnostic guidelines for PTSD, with most recent editions DSM-5 ICD-11 being best accepted. Generally, PTSD diagnosed according clusters symptoms occurring extreme stressors. Its pathogenesis multifactorial, including activation hypothalamic–pituitary–adrenal (HPA) axis, immune response, or even genetic discrepancy. The morphological alternation subcortical brain structures may also correlate symptoms. Prevention treatment methods vary from psychological interventions pharmacological medications. Overall, findings pertinent studies difficult generalize because heterogeneous patient groups, different events, criteria, designs. Future investigations needed determine which guideline inspection method early diagnosis strategies might prevent development

Abstract Posttraumatic stress disorder (PTSD) does not only have direct consequences for well-being, but it also comes with a significant risk severe somatic health consequences. A number of previous studies pointed to alterations in systems traumatized persons, as well the inflammatory system, which might be important links pathway between trauma, PTSD, and The aim this study was investigate acute responses PTSD patients compared healthy controls. Twenty-seven 15 controls were exposed Trier Social Stress Test (TSST), we measured salivary cortisol, alpha-amylase (sAA), plasma interleukin-6 (IL-6), heart rate variability (HRV) at different time points before, during after test. Results revealed similar controls, lower baseline cortisol levels higher IL-6 patients. Increases sAA significantly patients, while concentrations group intervention. HRV markedly decreased showed blunted response slower recovery TSST. These results confirm findings marked system dysregulations add literature on reactivity appears show system-specific changes. Overall, these implications our understanding potential resilience factors trauma.

Abstract Patients exposed to trauma often experience high rates of adverse post-traumatic neuropsychiatric sequelae (APNS). The biological mechanisms promoting APNS are currently unknown, but the microbiota-gut-brain axis offers an avenue understanding as well possibilities for intervention. Microbiome composition after exposure has been poorly examined regarding outcomes. We aimed determine whether gut microbiomes trauma-exposed emergency department patients who develop have dysfunctional microbiome profiles and discover potential associated mechanisms. performed metagenomic analysis on stool samples (n = 51) from a subset adults enrolled in Advancing Understanding RecOvery afteR traumA (AURORA) study. Two-, eight- twelve-week post-trauma outcomes stress disorder (PTSD) (PTSD checklist DSM-5), normalized depression scores (PROMIS Depression Short Form 8b) somatic symptom counts were collected. Generalized linear models created each outcome using microbial abundances relevant demographics. Mixed-effect random forest machine learning used identify associations between features encoded metabolic pathways metagenomics. Microbial species, including Flavonifractor plautii , Ruminococcus gnavus and, Bifidobacterium which prevalent commensal microbes, found be important predicting worse abundance data. Notably, through modeling pathways, highly predicted by decreased L-arginine related pathway genes increased citrulline ornithine pathways. Common species enriched individuals APNS. More notably, we identified mechanism reduces global arginine bioavailability, change that also demonstrated plasma with PTSD.

Observational studies suggest that posttraumatic stress disorder (PTSD) increases risk for various autoimmune diseases. Insights into shared biology and causal relationships between these diseases may inform intervention approaches to PTSD co-morbid conditions. We investigated the genetic contributions PTSD, 18 diseases, 3 immune/inflammatory biomarkers. Univariate MiXeR was used contrast architectures of phenotypes. Genetic correlations were estimated using linkage disequilibrium score regression. Bi-directional, two-sample Mendelian randomization (MR) performed independent, genome-wide significant single nucleotide polymorphisms; inverse variance weighted median MR estimates evaluated. Sensitivity analyses uncorrelated (MR PRESSO) correlated horizontal pleiotropy (CAUSE) also performed. considerably more polygenic (10,863 influential variants) than (median 255 variants).
