To address the limitations of traditional endotoxin adsorbents, which exhibit poor removal efficiency and low sample recovery when processing high-concentration samples, a novel cryogel, CG(HEMA-co-AM), based on acrylamide (AM) hydroxyethyl methacrylate (HEMA) as second monomer, was successfully designed synthesized. After optimizing epoxidation polymyxin B (PMB) conjugation processes, leading to successful preparation functionalized cryogel CG(HEMA-co-AM)@ECH@PMB, flow-through experiments showed that in Tris-HCl buffer at flow rate 6 mL/min, reached 99.82%, with maximum adsorption capacity 1408.38 EU/mg. In complex protein system containing BSA, HSA, Hb, LYS, OVA (each 10 mg/mL), 99.62%. Further investigations revealed pH ionic strength critically influenced regulation hydrophobic electrostatic interactions, thus significantly affecting efficiency. Additionally, weakly basic lysozyme exhibited higher rates compared strongly acidic proteins such BSA OVA. This integrates high excellent specificity features macroporous structure suitable for dynamic chromatographic separation. It offers novel, reusable adsorbent material preparation, biopharmaceutical production, clinical blood purification applications.
