Abstract Task-related neural activity is widespread across populations of neurons during goal-directed behaviors. However, little known about the synaptic reorganization and circuit mechanisms that lead to broad changes. Here we trained a subset in spiking network with strong interactions reproduce motor cortex decision-making task. activity, resembling data, emerged network, even untrained neurons. Analysis networks showed synapses, which were independent task determined dynamical state mediated spread task-related activity. Optogenetic perturbations suggest strongly-coupled, supporting applicability mechanism cortical networks. Our results reveal facilitates distributed representations task-variables by spreading from plastic entire through task-independent synapses.

Abstract State-of-the-art all-optical systems promise unprecedented access to neural activity in vivo , using multiphoton optogenetics allow simultaneous imaging and control of selected neurons at cellular resolution. However, achieve wide use stimulation imaging, simple strategies are needed robustly stably express opsins indicators the same cells. Here, we describe a bicistronic adeno-associated virus (AAV) that expresses both fast bright calcium indicator jGCaMP8s, soma-targeted (st) two-photon-activatable opsin, ChrimsonR. With this method, stChrimsonR with two-photon holography visual cortex mice drives robust spiking targeted cells, responses sensory stimuli spontaneous strong stable. Cells expressing construct show photostimulation similar measured from cells opsin via separate viruses. This approach is way prepare for imaging.

Abstract Sensory cortical representations can be highly dynamic, raising the question of how representational stability impacts learning. We train mice to discriminate number photostimulation pulses delivered opsin-expressing pyramidal neurons in layer 2/3 primary vibrissal somatosensory cortex. simultaneously track evoked neural activity across learning using volumetric two-photon calcium imaging. In well-trained animals, trial-to-trial fluctuations amount photostimulus-evoked predicted animal choice. Population levels declined rapidly training, with most active showing largest declines responsiveness. Mice learned at varied rates, some failing learn task time provided. The photoresponsive population showed greater instability both within and behavioral sessions among animals that failed learn. Animals also exhibited a faster deterioration stimulus decoding. Thus, response is associated sensory microstimulation task.

Neuroprosthetics offer great hope for motor-impaired patients. One obstacle is that fine motor control requires near-instantaneous, rich somatosensory feedback. Such distributed feedback may be recreated in a brain-machine interface using artificial stimulation across the cortical surface. Here, we hypothesized neuronal must contiguous its spatiotemporal dynamics to efficiently integrated by sensorimotor circuits.
